Relay Therapeutics Begins Phase 3 Trial of Zovegalisib, Triggering Milestone for X-Chem DEL Collaboration

Zovegalisib (搜索) is an inhibitor of phosphoinositide 3-kinase alpha (PI3K alpha (搜索)), an oncology target. Relay licensed the inhibitor after it emerged from an X-Chem (搜索) DEL screen. According to X-Chem, the molecule demonstrated differentiated pharmacology, mutant selectivity and a novel allosteric binding site, and it served as the starting point for Relay's development of zovegalisib, which is currently in clinical testing.
Collaboration terms and compound origins
Under the Research Collaboration and License Agreement, the initiation of the Phase 3 trial triggers a milestone payment to X-Chem (搜索). X-Chem did not disclose the financial terms of the milestone or details of the Phase 3 trial design, including endpoints, patient population or dosing.
"The unique selectivity and binding site of RLY-2608 are a testament to the power of the X-Chem (搜索) DEL platform to discover unique chemical matter with differentiated pharmacology," said X-Chem chief executive Karen Lackey. "We are thrilled that our discovery platforms helped provide a starting point for a potential new treatment for patients with breast cancer (搜索)."
Relay chief executive Sanjiv Patel pointed to the discovery work behind the program. "X-Chem (搜索)'s excellence in DEL technology was apparent from the very beginning of this exciting project, and we are pleased to continue to advance the clinical development of zovegalisib (搜索) in patients with PI3K alpha (搜索)-driven diseases," he said.

The zovegalisib (搜索) program is the second stage of a discovery-to-clinic path that began with a screening campaign rather than with a known chemical series. Relay has not disclosed the specific breast cancer (搜索) setting or the comparator for the Phase 3 study.
