Slate Medicines (搜索) has emerged from stealth mode with $130 million in Series A funding to develop a novel migraine prevention therapy that targets a different biological pathway than existing treatments. The newly formed biotech company announced Tuesday that it will use the investment from RA Capital Management (搜索), Forbion, Foresite Capital and another undisclosed backer to advance SLTE-1009 (搜索), a drug licensed from Guangzhou, China-based DartsBio Pharmaceuticals (搜索), into clinical testing.
The funding represents a significant bet on an alternative approach to migraine prevention at a time when many patients remain inadequately served by current therapies. More than 37 million people in the U.S. are estimated to live with migraines, which are pulsating headaches that can last hours to a few days and involve symptoms such as nausea and sensitivity to light and sound.
Targeting PACAP as Alternative to CGRP Inhibitors
SLTE-1009 (搜索) works by blocking PACAP (搜索) (pituitary adenylate cyclase-activating polypeptide), a protein involved in migraine onset that operates through different intracellular signaling pathways than the widely used CGRP (calcitonin gene-related peptide) inhibitors. This mechanistic distinction heightens expectations that drugs blocking PACAP might benefit people who aren't helped by CGRP inhibitors.
Over the last decade, drugs that block CGRP have emerged as a way to prevent or reduce the frequency of headaches. Several are sold by Amgen, Novartis, Pfizer, Eli Lilly, Lundbeck and Teva Pharmaceuticals, with some acquired through company buyouts. However, many patients don't respond to anti-CGRP medicines, leaving an opportunity for alternative approaches.
"PACAP (搜索) is a clinically validated target distinct from CGRP, offering potential benefit for patients underserved by current preventive therapies," Slate noted in its announcement.
Competitive Landscape and Delivery Advantages
Slate faces competition from Lundbeck, which holds the most advanced anti-PACAP (搜索) drug in development. Lundbeck's bocunebart (搜索), acquired through a 2019 acquisition of Alder BioPharmaceuticals, has showed promise in a pair of mid-stage trials. The Danish pharmaceutical company said earlier this month that it intends to work with U.S. regulators on the design of a Phase 3 program.
However, Slate's approach offers a potential convenience advantage. While bocunebart (搜索) is administered via intravenous infusion, SLTE-1009 (搜索) is designed as a subcutaneous injection that could be given at home. This delivery method could improve patient compliance and reduce healthcare system burden.
Leadership and Timeline
The company is led by Gregory Oakes, the former head of Landos Biopharma, which was acquired by AbbVie in 2024. Neil Buckley, a venture partner at RA Capital, serves as chief operating officer, while Chief Medical Officer Roger Cady previously held the position of vice president of neurology at both Alder and Lundbeck.
Slate, headquartered in Raleigh, North Carolina, expects to initiate Phase 1 testing of SLTE-1009 (搜索) in mid-2026. The company declined to provide additional details about the therapy's mechanism of action or preclinical data supporting its development.
