Solve Therapeutics (搜索), a clinical-stage biotechnology company developing next-generation antibody-drug conjugates (ADCs) for solid tumor malignancies, announced it has raised $120 million in an oversubscribed and upsized financing round. The funding will accelerate development of the company's clinical pipeline and proprietary CloakLink™ linker platform technology.
The round was led by Yosemite (搜索), with participation from new investors including Abingworth (搜索), Ally Bridge Group, B Capital, Balyasny Asset Management, Merck & Co. (搜索), and SymBiosis, alongside all existing investors. This latest funding follows a $75 million financing completed in December 2024, bringing Solve's total capital raised to $321 million since formation.
Addressing ADC Limitations with Novel Linker Technology
Solve Therapeutics (搜索) was founded to develop next-generation ADCs capable of addressing unique challenges presented by solid tumors. Traditional ADCs often face limitations related to payload hydrophobicity, including sub-optimal pharmacokinetics and plasma stability, which can compromise safety and efficacy.
The company's proprietary CloakLink™ technology was engineered to overcome these barriers by increasing ADC stability and decreasing ADC hydrophobicity across a broad range of drug-to-antibody ratios. According to CEO Dave Johnson, the ADCs built on this technology should stay fully intact when circulating through the bloodstream and only activate after reaching a tumor, enabling more precise delivery of toxic payloads.
"The result is a class of ADCs with improved pharmacokinetics, enhanced plasma stability, and reduced toxicity, enabling improved therapeutic indices and overall performance," the company stated.
Clinical Pipeline Progress
The company's lead programs, SLV-154 and SLV-324, are currently in Phase 1 clinical trials in patients with solid tumors. Both utilize the CloakLink™ platform and targeting antibodies that are specifically engineered for superior ADC performance. The ADCs are paired with novel diagnostic approaches to enable precision patient selection.
SLV-154 is being tested against head and neck cancer, non-small cell as well as small cell lung cancers, and bladder tumors, among others. The company has not yet disclosed which cancers SLV-324 will target.
Johnson indicated that the trials are enrolling ahead of schedule, stating: "We're excited about the progress across our clinical programs and look forward to sharing more as the data emerges." The data generated so far, while early, are "validating and consistent" with what the company observed in preclinical testing.
