The MFDS (search) approved lenacapavir, marketed as Sunlenca Injection and Sunlenca Tablets, for treating human immunodeficiency virus type 1 (HIV-1 (search)) infection in adult patients with multidrug-resistant disease. This first-in-class treatment represents a novel therapeutic approach by targeting the capsid protein (search) of HIV-1.
Lenacapavir works through a unique mechanism that blocks the process through which the virus enters the cell nucleus, while simultaneously inhibiting viral assembly and release and inducing abnormal capsid formation to prevent replication. The approved indication specifically covers adult patients with multidrug-resistant HIV-1 (search) infection who are difficult to treat with existing antiretroviral therapies and have failed prior treatments with limited alternative options.
"This approval is expected to provide a new treatment opportunity that addresses unmet medical needs not fulfilled by existing therapies," the MFDS (search) stated. HIV infection without proper treatment can severely compromise immune function and progress to acquired immunodeficiency syndrome (AIDS (search)), with multidrug-resistant patients facing particularly limited treatment options.
HER2-Targeted Therapy for Rare Lung Cancer Subtype
The ministry also approved sevabertinib (search) (Hernexeos (search)) as an orphan drug for patients with locally advanced or metastatic non-squamous non-small cell lung cancer (search) harboring HER2 (search) gene mutations who have received prior systemic therapy. This oral tyrosine kinase inhibitor specifically targets HER2, addressing a patient population with limited treatment alternatives.
When HER2 (search) mutations occur, signals for cell proliferation and survival become excessively activated. Sevabertinib (search) works by blocking this signal transduction to suppress cancer cell growth, providing precision targeted therapy for this specific genetic subtype.
Fast-Track Regulatory Pathway Accelerates Access
The lung cancer treatment was reviewed under Korea's Global Innovative Products on Fast Track (GIFT) program, representing the 43rd case designated under this initiative. The program shortens review periods for highly innovative medicines among treatments for life-threatening serious and rare diseases to support rapid market entry.
"We will continue to pursue swift review and approval of innovative medicines based on regulatory science expertise," a ministry official said. The ministry emphasized its commitment to supplying treatments with confirmed safety and efficacy more quickly through the fast-track review system and expanding treatment opportunities for patients with rare and intractable diseases.
