Novel Gene-Agnostic Approach
SPVN06 distinguishes itself from existing gene-specific therapies through its mutation-agnostic design. The AAV-based gene therapy is engineered to preserve vision by protecting cone photoreceptors (search)—the light-sensing cells responsible for visual acuity and color vision—through neuroprotective effects on cone metabolism.

Together, these elements aim to slow or stop the degeneration of cone photoreceptors (search) that inevitably leads to blindness in patients with rod-cone dystrophies (search).
Broad Therapeutic Potential
The gene-agnostic approach positions SPVN06 for potential applications across multiple retinal conditions where cone photoreceptor loss leads to blindness. Beyond inherited retinal diseases (search) like RP, the therapy shows promise for non-inherited conditions such as age-related macular degeneration (search) (AMD) and Geographic Atrophy (search) secondary to dry AMD.

Trial Design and Regulatory Strategy
The PRODYGY trial was conducted in two phases. In the dose-escalation portion (Step 1), nine patients received SPVN06 across three different dose levels. The second phase (Step 2) randomized 24 patients into three study arms: high dose (n=9), medium dose (n=9), or an untreated control group (n=6).
The trial is designed to assess safety and exploratory efficacy endpoints, including functional, anatomical, and quality of life measures. Primary endpoint analysis will occur after all patients have completed at least one year of follow-up.
