Trial Design and Patient Population
The Phase 2a study (NCT06427668) evaluated Tazbentetol (搜索) in participants with mild-to-moderate Alzheimer's disease (搜索), defined by standardized mini mental status exam (MMSE) scores between 16 and 26. Patients received either 300mg or 150mg of Tazbentetol daily for up to 24 weeks, beginning with a four-week placebo-controlled period. The study's primary endpoints included SMMSE, Clinical Dementia (搜索) Rating Sum of Boxes (CDR-SB), Activities of Daily Living (ADL), and neurophysiological EEG biomarker measures of AD-related brain activity.
Rapid Cognitive Benefits
Results for the 300mg dose, presented at the Clinical Trials on Alzheimer's Disease (搜索) (CTAD) conference on December 4, 2025, demonstrated rapid cognitive gains. Participants displayed evidence of early cognitive benefits, with a >2.5 point average increase in SMMSE within four weeks versus placebo (p < 0.05).
The cognitive improvements proved durable, with sustained improvement observed through the 24-week open-label period across SMMSE, CDR-SB, and ADL outcomes. Notably, scores on the CDR-SB and ADL outcome measures continued to improve over baseline through 40 weeks, suggesting increasing benefit over time.
Neurophysiological Evidence of Synaptic Regeneration
Treatment with 300mg Tazbentetol (搜索) was associated with significant improvement in AD-related brain activity patterns as assessed by EEG. The analysis revealed a reversal of established metrics of cortical "slowing" across multiple frequency bands, providing objective neurophysiological biomarker evidence of synaptic regeneration.

Safety Profile
Tazbentetol (搜索) demonstrated a favorable safety profile with no severe or treatment-related adverse events. The therapy was well tolerated by participants taking the drug both as a monotherapy and in combination with standard of care therapy.
