Strand Therapeutics (搜索) announced the completion of a $153 million Series B financing round on Thursday, bringing the Massachusetts-based biotech's total funding to over $250 million as it advances its pipeline of programmable mRNA therapies for cancer treatment. The round was led by Kinnevik (搜索), with new investors including Regeneron Ventures, ICONIQ, Amgen Ventures, and continued participation from existing backers Eli Lilly and Company.
The funding will primarily support the advancement of Strand's lead program STX-001, an investigational self-replicating mRNA therapy that encodes the immunomodulatory protein interleukin-12 (IL-12 (搜索)). Unlike traditional mRNA therapies, STX-001 is delivered directly into tumors to reshape the microenvironment and trigger a systemic anti-cancer immune response.
Clinical Progress Shows Promise in Treatment-Resistant Patients
STX-001 is currently being evaluated in an ongoing first-in-human Phase I study for patients with solid tumors that are refractory to immune checkpoint inhibitors. The company recently presented initial clinical data at the 2025 ASCO Annual Meeting, demonstrating multiple RECIST responses including cases of complete response and complete metabolic response.

"We have observed systemic immune activation and anti-tumor responses, including responses in non-injected lesions, across multiple tumor types."
The therapy has shown signals of treatment response with several partial responses and a confirmed case of complete response, along with multiple cases of prolonged disease stabilization and a favorable safety profile in treatment-resistant patients.
Next-Generation Platform Technology
Strand's proprietary platform distinguishes itself through the use of self-replicating mRNA combined with programmable genetic circuits. These "logic circuits" enable precise control over the location, timing, and degree of therapeutic expression, allowing for controlled delivery of the therapeutic payload.
IL-12 (搜索), the cytokine encoded by STX-001, is a potent pro-inflammatory molecule primarily produced by antigen-presenting cells such as macrophages and dendritic cells. The cytokine holds significant promise in cancer immunotherapy due to its robust immunostimulatory effects, with Strand's approach designed to reprogram the tumor microenvironment while ensuring localized and durable therapeutic activity.
Pipeline Expansion with Systemic Delivery
Beyond STX-001, Strand is developing STX-003, which the company describes as a "world-first systemically administrable mRNA therapy with tumor targeting." This candidate is programmed to avoid off-target payload delivery, including liver avoidance, and similarly carries an IL-12 (搜索)-expressing payload using self-replicating mRNA technology.
Preclinical data for STX-003 presented at the 2025 AACR and ASGCT annual meetings demonstrated the candidate's potential to target IL-12 (搜索) expression to tumors following systemic administration, creating an effective anti-tumor therapy that was well tolerated. The company expects to begin dosing STX-003 in patients next year.
Investment Validation and Future Outlook

The company's platform combines engineered next-generation mRNA modalities with genetically programmed logic circuits, positioning it to develop scalable and accessible targeted therapies for cancer and other chronic diseases.
The funding comes at a time when the broader mRNA field faces some policy headwinds, with Health Secretary Robert F. Kennedy Jr. recently canceling $500 million worth of mRNA vaccine contracts under the Biomedical Advanced Research and Development Authority, though his actions focused specifically on respiratory infection vaccines rather than cancer therapeutics.
