Surrozen Advances Wnt-Based Retinal Therapeutics Pipeline, Plans IND Filing for SZN-8141 in Second Half of 2026
核心洞察
Surrozen (搜索) is developing next-generation ophthalmology therapeutics targeting Wnt signaling pathways to treat sight-threatening retinal diseases including diabetic macular edema (搜索) and wet AMD (搜索).
The company plans to submit an Investigational New Drug application for lead candidate SZN-8141 to the FDA in the second half of 2026, marking a key regulatory milestone.
Boehringer Ingelheim achieved a research milestone for partnered asset SZN-413, triggering a $5 million milestone payment and reflecting positive outcomes from IND-enabling toxicology studies.
Surrozen (搜索) Inc. is advancing its pipeline of Wnt-based therapeutics for retinal diseases, with plans to submit an Investigational New Drug (IND) application for its lead candidate SZN-8141 to the FDA in the second half of 2026. The South San Francisco-based biotechnology company is pioneering targeted therapeutics that harness Wnt signaling to address underlying drivers of sight-threatening ophthalmic conditions.
The company's approach centers on developing multifunctional biologics that selectively activate Wnt signaling in combination with other key disease pathways, potentially offering advantages over current standard-of-care treatments. "2025 was a year of significant momentum as we progressed two lead product candidates in retinal vascular disease, advanced our platform for developing multifunctional biologic candidates that selectively activate Wnt signaling in combination with other key disease pathways," said Craig Parker, President and Chief Executive Officer of Surrozen (搜索).
Lead Pipeline Assets Show Promise in Preclinical Models
SZN-8141 is being developed for the treatment of diabetic macular edema (搜索) (DME) and neovascular age-related macular degeneration (搜索) (wet AMD (搜索)). The therapeutic combines Frizzled 4 (搜索) (Fzd4) agonism and vascular endothelial growth factor (VEGF (搜索)) antagonism, potentially providing benefits over treatment with single agents. Data generated in preclinical models of retinopathy demonstrated that SZN-8141 stimulated Wnt signaling and induced normal retinal vessel regrowth while suppressing pathological vessel growth.
The company's second lead candidate, SZN-8143, is being developed for DME, wet AMD (搜索), and uveitic macular edema (搜索) (UME). This therapeutic combines Fzd4 agonism, VEGF (搜索) antagonism, and interleukin-6 (IL-6) antagonism. Preclinical data showed that SZN-8143 stimulated Wnt signaling and induced normal retinal vessel regrowth while suppressing pathological vessel growth.
Both candidates represent potential improvements over the current standard of care for diabetic retinopathy (搜索), retinal vein occlusion (搜索) and wet AMD (搜索), which involves intravitreal administration of anti-VEGF (搜索) therapies, including monotherapies and dual-pathway agents targeting VEGF and Ang-2 (搜索).
Partnership Milestone Validates Platform Technology
In March 2026, Boehringer Ingelheim achieved a research milestone under its collaboration agreement for SZN-413, reflecting positive outcomes from IND-enabling GLP toxicology studies. This milestone triggered a $5.0 million payment to Surrozen (搜索), validating the company's SWAP™ technology platform.
SZN-413 is a bi-specific antibody targeting Fzd4-mediated Wnt signaling designed using Surrozen (搜索)'s proprietary technology. Data generated by Surrozen with SZN-413 in preclinical models of retinopathy demonstrated that the therapeutic potently stimulated Wnt signaling in the eye, induced normal retinal vessel regrowth, suppressed pathological vessel growth and reduced vascular leakage.
Under the partnership terms, Boehringer Ingelheim received an exclusive, worldwide license to develop SZN-413 and other Fzd4-specific Wnt-modulating molecules in exchange for an upfront payment of $12.5 million and up to $586.5 million in success-based development, regulatory, and commercial milestone payments, plus mid-single digit to low-double digit royalties on sales.
Scientific Presentations and Leadership Strengthening
Surrozen (搜索) plans to present retinal vascular research on SZN-8141 at the 2026 Association for Research in Vision and Ophthalmology (ARVO) Annual Meeting on May 4, 2026. The poster presentation is titled "Wnt signaling activation synergizing with VEGF (搜索) inhibition in preclinical models of retinal vascular disease."
The company has strengthened its strategic leadership and ophthalmology expertise by forming a Clinical Advisory Board composed of retinal disease experts and appointing industry veterans Daniel Chao, M.D., Ph.D., as Vice President and Head of Clinical Development and Andrew Maleki as Chief Financial Officer.
Financial Position Supports Development Plans
As of December 31, 2025, Surrozen (搜索) reported cash and cash equivalents of $89.2 million, compared to $34.6 million as of December 31, 2024. The company received additional funding through net proceeds of $26.9 million from sales of common stock under its at-the-market sales program in January 2026 and $3.3 million from warrant exercises in February and March 2026.
Research and development expenses increased to $29.4 million for the year ended December 31, 2025, compared to $21.1 million for 2024, primarily reflecting increased manufacturing costs, lab expenses and consulting fees for ophthalmology programs. The company discontinued clinical development of SZN-043, resulting in decreased clinical expenses that partially offset the increases in other R&D areas.
