Tolerance Bio (搜索), Inc. has signed an exclusive license agreement with Tanabe Pharma (搜索) Corporation for MT-2990, a clinical-stage anti-IL-33 (搜索) monoclonal antibody now renamed TLB-33 (搜索), granting the Philadelphia-based biotechnology company worldwide development rights and exclusive commercialization rights outside Japan. Tanabe Pharma retains commercialization rights in Japan.

Clinical Data and Planned Development
TLB-33 (搜索) is a human immunoglobulin G1 (IgG1) monoclonal antibody that binds to and neutralizes interleukin-33 (IL-33 (搜索)). Tanabe Pharma (搜索) has already evaluated the asset in over 150 subjects across five clinical studies, generating safety, tolerability, pharmacologic and clinical data that Tolerance Bio (搜索) says support development in new indications.
Tolerance Bio (搜索) is preparing to advance TLB-33 (搜索) into Phase 2 clinical development beginning in 2027 to evaluate its potential to preserve thymic function and enhance immune resilience, with additional immune indications also under consideration.

Leon added that IL-33 (搜索) signaling has been implicated in thymic involution (搜索) in inflammatory settings, and that blocking IL-33 may offer an opportunity to preserve thymic function and improve immune resilience.
Rationale: IL-33 and Thymic Involution
IL-33 (搜索) is an alarmin released upon cellular stress or tissue injury that drives activation of innate and adaptive immune responses. Dysregulated IL-33 signaling has been implicated in inflammatory pathways believed to contribute to thymic dysfunction and the progressive decline of immune function with age, as well as acute thymic involution (搜索) in certain immune conditions.
The thymus plays a central role in T-cell maturation and immune competence but progressively involutes with age and in the context of acute inflammatory triggers. According to Tolerance Bio (搜索), this decline contributes to increased susceptibility to infections, cancer, autoimmunity and reduced vaccine responsiveness.
A Multi-Modality Pipeline Around Thymic Biology
TLB-33 (搜索) expands a pipeline that Tolerance Bio (搜索) has built across biologics, mRNA and cell therapies targeting thymus biology. The company is among the first to specifically target thymic biology as a therapeutic strategy to preserve immune resilience.
The license follows a recently announced agreement with NeoImmuneTech (搜索) for the long-acting interleukin 7 (IL-7) fusion protein efineptakin alfa, a deal valued at up to $260 million. That agreement was underpinned by evidence that IL-7 stimulates thymic function and T-cell differentiation, proliferation and survival.
"Together with our recently licensed long-acting IL-7 program, TLB-33 (搜索) gives us complementary approaches to preserving and restoring thymic function," Leon said.
Tolerance Bio (搜索) launched in October 2024 with $17.2 million in seed funding and is backed by life-sciences investors including Columbus Venture Partners, Criteria Bio Ventures, Pacific 8 Ventures and BioAdvance Capital. The company expects to announce the completion of a financing round to support advancement of TLB-33 (搜索) and efineptakin alfa through planned clinical trials.
