Valneva SE (search) announced positive results from its Phase 1 clinical trial of VLA1601, a second-generation adjuvanted inactivated vaccine candidate against the Zika virus (search), demonstrating significantly enhanced immune responses compared to the company's first-generation candidate.
Enhanced Immunogenicity Profile
The randomized controlled Phase 1 trial, designated VLA1601-102, enrolled approximately 150 participants aged 18 to 49 years in the United States. Participants received two administrations of VLA1601 four weeks apart, testing low, medium, and high doses of the highly purified inactivated aluminum-adjuvanted vaccine candidate.
The study also evaluated the low dose of VLA1601 with additional adjuvants, including CpG 1018® adjuvant from Dynavax Technologies Corporation and 3M-052-AF (search) adjuvant from the Access to Advanced Health Institute (search) (AAHI).
Data collected up to Day 57 showed that the strongest immune response occurred in the double-adjuvant treatment arms (Low+alum+3M-052-AF (search) and Low+alum+CpG1018), which demonstrated statistically significantly higher neutralizing antibody titers at Day 43 and Day 57 compared to the single-adjuvant treatment arm.
Superior Performance Over First-Generation Candidate
The immune response induced by the double-adjuvanted VLA1601 second-generation vaccine candidate showed marked improvement over the first-generation vaccine candidate, with higher peak seroconversion rates exceeding 93% versus 86% and peak Geometric Mean Fold Increase of titers greater than 56-fold versus greater than 7-fold. Phase 1 results from Valneva's first-generation Zika vaccine candidate were reported in 2018.
Safety Profile and Manufacturing Platform
VLA1601 demonstrated a generally safe and well-tolerated profile across all five treatment arms, with no safety concerns identified. An independent Data Safety Monitoring Board confirmed no safety issues during the trial period.
The vaccine candidate is developed on the original manufacturing platform of Valneva's licensed Japanese encephalitis vaccine IXIARO®, which was further optimized to develop the company's inactivated, adjuvanted COVID-19 vaccine VLA2001, the first COVID-19 vaccine to receive standard marketing authorization in Europe.
Global Health Context
According to the World Health Organization, there is scientific consensus that Zika virus (search) infection can cause congenital microcephaly (search) and Guillain-Barré syndrome (search). Since 2013, 31 countries and territories have reported cases of congenital microcephaly and other central nervous system malformations associated with Zika virus infection.
The Zika virus (search) is currently circulating in Mexico, Central and South America, many Caribbean countries and territories, and geographically limited areas of the continental United States. To date, 89 countries and territories have reported evidence of mosquito-borne Zika virus transmission, though global surveillance remains limited.

Despite the demonstrated medical need and positive trial results, regulatory pathways and market opportunities for potential Zika vaccines remain uncertain. Valneva indicated it will only consider further potential development steps for VLA1601 if concrete major private and public funding opportunities materialize.
