Vedana Therapeutics Launches with $46M Series A to Develop Next-Generation PACAP-Targeting Migraine Therapies

The company is developing antibody drugs targeting PACAP (搜索), a nervous system protein that drives migraines in a similar but distinct way from CGRP, aiming to fill a treatment gap where over half of patients don't respond to existing CGRP-blocking therapies.
Vedana's leadership team includes veterans who discovered and developed multiple approved migraine (搜索) therapies, including CGRP antibodies and the emerging PACAP (搜索) drug class at Alder BioPharmaceuticals and Labrys Biologics.
Vedana Therapeutics (搜索), a biotechnology startup built by the scientists and executives behind multiple approved migraine (搜索) therapies, emerged from stealth Wednesday with $46 million in Series A financing to develop next-generation preventive treatments targeting the pituitary adenylate cyclase-activating polypeptide (PACAP (搜索)) pathway.
The financing was co-led by Westlake BioPartners (搜索) and Canaan Partners (搜索), with participation from Dawn Biopharma and Alexandria Venture Investments. The company plans to use the proceeds to advance its portfolio of internally discovered, subcutaneously delivered antibody therapies into human trials, which are on track to begin next year.
Addressing the CGRP Treatment Gap
The first generation of specialized migraine (搜索) therapies — monoclonal antibodies targeting calcitonin gene-related peptide (CGRP) — represented a major therapeutic advance when they reached the market toward the end of the last decade. These medicines work by inhibiting CGRP, a protein that transmits pain signals, widens blood vessels, and triggers inflammation in tissues surrounding the brain.
Yet a substantial proportion of patients remain inadequately treated. By some estimates, more than half of patients do not respond to or discontinue CGRP-blocking therapies. Vedana cites data indicating that two-thirds of migraine (搜索) patients are still unable to gain optimal control of their condition.
"First-generation CGRP therapies were a major step forward in the treatment of migraine (搜索), but most patients are still not adequately controlled," said Anurag Agarwal, Ph.D., co-founder and chief executive officer of Vedana. "We are now focused on what comes next — targeting biology beyond CGRP, like PACAP (搜索) and combinations thereof, to deliver on our vision of migraine freedom for all patients."
The PACAP (搜索) Opportunity and Competitive Landscape
PACAP (搜索), like CGRP, plays a critical role in migraine (搜索) physiology and is emerging as a complementary approach to treat patients. Vedana is not alone in pursuing this target. The Danish company Lundbeck, through its $2 billion acquisition of Alder BioPharmaceuticals in 2019, gained an anti-PACAP drug that has succeeded in multiple mid-stage clinical trials. Two other developers — Mentari Therapeutics, which announced plans in May to go public through a reverse merger, and Slate Medicines, a newly formed biotech that has raised $130 million from firms including RA Capital Management, Forbion, and Foresite Capital — are also advancing PACAP programs.
Agarwal argues that Vedana's biggest differentiator is its leadership team. Co-founder and chief scientific officer Leon Garcia, Ph.D., oversaw discovery and development of both anti-CGRP and anti-PACAP (搜索) antibodies at Alder BioPharmaceuticals. Chief medical officer Ernesto Aycardi, M.D., managed the pivotal and post-marketing studies for Teva Pharmaceuticals' CGRP product AJOVY (fremanezumab-vfrm).
"These are the people that have done it before, both for CGRP and PACAP (搜索). They understand the biology," Agarwal said. "They are the people who've gone from discovering a drug to delivering it to the migraine (搜索) patient."
A Team Built on Migraine (搜索) Drug Development Experience
Vedana's board and advisors include an extensive roster of industry veterans. Rob Lenz, M.D., Ph.D., who served as head of global development at Amgen during the advancement of Aimovig (erenumab-aooe), serves as executive chair of the board. Marcelo Bigal, M.D., Ph.D., former CMO of Labrys Biologics and head of development at Teva, and Steven James, former CEO of Labrys, both sit on the board. The company has also recruited leading academic researchers, including Messoud Ashina, M.D., Ph.D., director of the Migraine (搜索) Unit at the University of Copenhagen, and David Dodick, M.D., a globally recognized migraine expert.

"Importantly, they bring together the lived experiences from multiple migraine companies and medicines in large and small biotech. They are focused entirely on what the next generation of medicines needs to deliver for patients."
Dual-Program Pipeline Strategy
Vedana has built a portfolio of internally discovered, subcutaneously delivered long-acting antibodies designed for self-injection in the convenience of patients' homes. The lead program is a next-generation anti-PACAP (搜索) antibody, offering a novel mechanism beyond currently approved drugs. The second program is a bispecific antibody targeting both PACAP and CGRP, an approach that could potentially deliver additional efficacy for patients who do not achieve relief with monotherapies.
"Drawing on that experience, we designed the next-generation of highly potent long half-life antibodies, targeting self-injecting options for patients in the convenience of their home," said Leon Garcia.
