Ventyx Biosciences announced significant expansion of its Phase 2 clinical program for VTX2735, a peripherally restricted NLRP3 (搜索) inhibitor being developed for recurrent pericarditis (搜索), alongside strategic additions to its advisory board. The clinical-stage biopharmaceutical company is extending its ongoing multicenter, open-label Phase 2 study into Canada, the European Union, and the United Kingdom to evaluate once-daily dose ranging in preparation for global Phase 3 development.
The company has received Health Canada regulatory approval to activate clinical sites by the end of December and has initiated the Clinical Trial Application filing process in the EU and UK. Starting in December, the study will switch to a once-daily dosing regimen, with the primary endpoint for these cohorts still assessed at week 6, but the extension period will now extend through 24 weeks compared to the current 13-week extension.
Strategic Advisory Board Expansion
Ventyx strengthened its leadership team with two notable additions. Mark McKenna, MBA, joins as Strategic Advisor, bringing extensive experience as founder, Chairman and CEO of Mirador Therapeutics (搜索), a precision medicine company focused on immunology and inflammation. McKenna previously served as Chairman, President and CEO of Prometheus Biosciences (搜索) from 2019-2023, where the company created the first precision therapeutics for immune-mediated diseases before being acquired by Merck (搜索) for $10.8 billion in June 2023.
Dr. Peter Libby, MD, joins as Clinical Advisor, bringing cardiovascular expertise from his role as a specialist at Mass General Brigham Heart & Vascular Institute and immediate past president of the International Atherosclerosis (搜索) Society. His research focuses on the role of inflammation in vascular diseases, such as atherosclerosis. Notably, Dr. Libby instigated and helped lead the Canakinumab Anti-Inflammatory Thrombosis Outcomes Trial (CANTOS) that provided clinical validation of the role of inflammation in atherosclerosis.

Current Phase 2 Trial Design and Timeline
VTX2735 is currently being evaluated in an ongoing, multicenter, open-label Phase 2 study in patients with severe recurrent pericarditis (搜索) (NCT06836232). The study is evaluating a 150 mg twice-daily dosing regimen with the primary endpoint measured at week 6. Key endpoints include safety, change in the numerical rating scale (NRS) pain score, and change in high sensitivity C-reactive protein (hsCRP).
The company revised its guidance for topline data release from the interim analysis of the Phase 2 recurrent pericarditis (搜索) trial to Q1 2026. "While the timing represents a modest shift from our prior guidance it also provides us with an opportunity to introduce dose-ranging studies with our new once-daily or QD formulation in the current Phase 2 study while also expanding into Canada, EU and the UK, a strategy we feel will accelerate Phase 3 timelines," added Dr. Mohan.

Recurrent pericarditis (搜索) is a condition in which the pericardium, a double-layered sac that surrounds and protects the heart, is inflamed and subsequent flaring leads to severe pain and heart complications. The NLRP3 (搜索) inflammasome is believed to be a major instigator of the aberrant immune response in the pericardium of recurrent pericarditis patients. By targeting NLRP3, VTX2735 has the potential to treat patients experiencing an active flare and prevent future recurrences, streamlining the treatment for patients with recurrent pericarditis.
