Verastem Oncology Restructures Leadership to Advance KRAS-Targeted Cancer Pipeline

Verastem Oncology announced strategic leadership changes designed to accelerate its next phase of growth, appointing Michael Kauffman, M.D., Ph.D., as president of development and John Johnson as chairman of the board. The moves come as the company progresses its commercial launch of AVMAPKI (search) FAKZYNJA (search) CO-PACK and advances its pipeline of RAS/MAPK pathway-targeted cancer therapies.
Kauffman, who has served as lead director since 2016, will transition from the board to join the executive leadership team full-time. Johnson, a board member since 2020, succeeds Kauffman as chairman. As part of the restructuring, chief operating officer Matthew Ros will depart as the company streamlines its operational structure.
Leadership Transition Reflects Strategic Priorities

Johnson brings more than three decades of experience across corporate strategy, operations, and oncology drug commercialization. He previously served as Company Group Chairman of Biopharmaceuticals at Johnson & Johnson and as President of Eli Lilly's Worldwide Oncology unit. "Verastem is at a pivotal moment with the initial successful launch of AVMAPKI (search) FAKZYNJA (search) CO-PACK, which has provided a benefit to women where previously there were no FDA-approved treatments specifically for their disease," Johnson said.
Focus on KRAS G12D Inhibitor Development
Kauffman expressed particular enthusiasm about VS-7375, describing it as "a once-in-a-lifetime opportunity." The potential best-in-class KRAS G12D (search) dual ON/OFF inhibitor "could transform outcomes for patients with currently limited options," he said. Kauffman's background includes co-founding and leading Karyopharm (search) through the global approvals of XPOVIO and serving as Chief Medical Officer at Onyx Pharma during the development of Kyprolis.
RAMP 301 Trial Completes Additional Enrollment
The company also announced completion of additional patient enrollment for RAMP 301, its international Phase 3 confirmatory trial in recurrent low-grade serous ovarian cancer (LGSOC). Following a pre-planned interim analysis, the Independent Data Monitoring Committee recommended a modest one-time increase of 29 patients across KRAS (search) mutation status. The company remains blinded to the interim analysis results.
RAMP 301 is evaluating the combination of avutometinib plus defactinib versus standard chemotherapy for patients with recurrent LGSOC with and without a KRAS (search) mutation. The trial serves as a confirmatory study for the initial indication and has the potential to expand the indication regardless of KRAS mutation status. Verastem expects to report topline results of the primary endpoint in mid-2027.
Commercial Foundation with AVMAPKI FAKZYNJA CO-PACK
The FDA approved AVMAPKI (search) FAKZYNJA (search) CO-PACK on May 8, 2025, for adult patients with KRAS (search)-mutated recurrent LGSOC who have received prior systemic therapy. The approval was granted under accelerated approval based on tumor response rate and duration of response, with continued approval contingent upon verification of clinical benefit in a confirmatory trial.
The combination therapy targets the RAS/MAPK pathway through complementary mechanisms. AVMAPKI (search) (avutometinib) inhibits MEK (search) kinase activity while blocking compensatory reactivation of MEK by upstream RAF (search). FAKZYNJA (search) (defactinib) is a FAK (search) inhibitor that addresses drug resistance mechanisms activated when RAF and MEK are blocked.
Expanding Pipeline Applications
Beyond LGSOC, Verastem is evaluating avutometinib plus defactinib in combination with standard-of-care chemotherapy for first-line treatment of advanced pancreatic cancer (RAMP 205) and advanced KRAS (search) G12C mutant non-small cell lung cancer (RAMP 203). These investigational uses are not yet approved by regulatory authorities.
The leadership changes position Verastem to leverage its commercial success in rare ovarian cancer while advancing its broader pipeline of RAS/MAPK pathway inhibitors, with particular emphasis on the KRAS G12D (search) inhibitor VS-7375 that represents a significant portion of the company's future growth strategy.
