PTN primarily focuses on off-patent, or generic, drugs that lack sufficient data for pediatric populations. Because the exclusive rights to market these drugs have expired, pharmaceutical companies often do not invest in testing them in children, leaving healthcare providers with limited information on safe and effective usage.
"Prior to 2022, we did not know the correct dosing for >90% of medicines used in children; now that is dramatically different because of the work of PTN," said Dr. Danny Benjamin, Principal Investigator and Chair of the Pediatric Trials Network (search).
Impact on Drug Labeling and Clinical Practice
Data from PTN trials are instrumental in helping regulatory bodies revise drug labels, ensuring safer and more effective use of medications in infants and children. To date, these efforts have resulted in 23 label changes by the Food and Drug Administration (FDA), significantly improving health outcomes for young patients.
Studies on Breastfeeding Mothers
PTN's work extends to vulnerable populations such as breastfeeding mothers. The CUDDLE study, for example, has significantly enhanced drug safety data for this group. Historically, there has been limited information on the effects of drugs on breast milk, causing uncertainty for new mothers. PTN is currently researching 30 common medications used by breastfeeding women to provide crucial safety information.
Optimizing Treatments for Critically Ill Children
For critically ill children requiring specialized care, PTN's research is crucial in optimizing treatments like Extracorporeal Membrane Oxygenation (ECMO) for severe heart and lung conditions in neonates and children. PTN's research ensures the safe and effective dosing of drugs for these vulnerable patients.
Clinician Feedback and Future Directions
Physicians nationwide have reported that PTN's work has significantly informed their practices and saved patients' lives. "In our own practice as physicians who provide clinical care to infants and children, we prescribe medications that have been studied by PTN every day," noted Dr. Rachel Greenberg, Chair of the Pediatric Trials Network (search) Steering Committee.
Looking ahead, PTN plans to expand its efforts in enhancing pediatric health through ongoing research, particularly with new health challenges emerging. Increased funding would enable larger trials to support clinicians' critical decisions and improve outcomes for future generations.
