"Only about 30 to 40% of dogs (with osteosarcoma (搜索)) will survive one year" with standard care, Mamula explained. "Adding our therapy to that standard of care, survival now increases from 30 to 40% up 60 or 70%."
The vaccine's effectiveness was demonstrated in the case of Ranger, a 7-year-old golden retriever diagnosed with osteosarcoma (搜索) in March 2019. After exhausting conventional treatments including amputation, chemotherapy, and experimental drugs, owner Rick Kneisel turned to Mamula's vaccine as a last resort. Within a month of receiving two doses in November 2019, Ranger's tumor had stopped growing, and within three months, the cancer (搜索) no longer appeared on X-ray scans.
Targeting Cancer at the Molecular Level
The immunotherapy is designed to target the EGFR (搜索)/HER2 (搜索) family of proteins that enable cancerous tumor cells to grow and spread. The vaccine works by blocking the signals these mutant proteins send to cells, instructing them to multiply and spread throughout the body.
Beyond disrupting tumor growth signals, the vaccine also prompts the production of antibodies capable of killing tumor cells and pathogens. This "very strong" immune response, driven by the vaccine's precise targeting of tumor sites, distinguishes this treatment from other available therapies, according to Mamula.
Clinical Trial Structure and Accessibility
The ongoing clinical trials follow an "open-label" design, where both patients and providers are aware of the treatment being administered. Dogs enrolled in the study receive two doses of the vaccine three weeks apart and are monitored closely throughout the process.
The treatment is currently offered free to pet owners as part of the clinical trials and is open to dogs with certain forms of cancer (搜索). Veterinarians travel with mobile units, meeting pet owners in parking lots across New York and Connecticut, with some owners traveling from as far as New Hampshire and Philadelphia for treatment.
Market Potential and Regulatory Timeline
The vaccine's potential impact could be substantial given the scope of canine cancer (搜索). Statistics show that one in four dogs will develop cancer in their lifetime, with larger breeds facing greater risk. Among dogs aged 10 and older, that risk increases to one in two. With approximately 90 million dogs living in the United States in 2024, according to the American Veterinary Medical Association, the addressable population is significant.
"You start doing the numbers with 90 million dogs, there are large populations of dogs in this country that are destined to get cancers," Mamula noted.
Researchers are "hopeful" that the U.S. Department of Agriculture, which regulates veterinary vaccines, will license the treatment within the next six months. Once approved, TheraJan (搜索) plans to manufacture and distribute the vaccine to retail locations and veterinary clinics across the United States.
While pricing has not been finalized, Mamula indicated the vaccine will "very likely be the least expensive cancer (搜索) therapy a dog can get."
Personal Motivation Behind the Research
Mamula's dedication to canine cancer (搜索) research stems from personal experience. Approximately twelve years ago, his 10-year-old golden retriever, Savannah, died from hemangiosarcoma (搜索), an aggressive cancer arising from blood vessels.
"I realized at the time that the tools the veterinarian oncologists have at their disposal were just not what one can find in treating human cancers by comparison," he reflected.
This loss motivated Mamula to partner with Gerald Post, then a Norwalk-based veterinary oncologist, in 2015 to launch early-phase studies for the novel cancer (搜索) vaccine. This initial work "paved the way" for the current clinical trials, though Post is no longer involved in the research.
Since the initial study, Mamula and his team have refined their understanding of which dog breeds and cancer (搜索) types respond most effectively to the vaccine, allowing them to optimize their therapeutic approach.
Mamula describes this cancer (搜索) vaccine research as "the most satisfying work that I've done" in his more than 35 years at Yale, often receiving updates from dog owners years after their pets receive treatment, sharing stories of their companions' continued health and happiness.
