Zepbound Receives Health Canada Approval as First Dual-Receptor Obesity Treatment
核心洞察
Zepbound (tirzepatide injection) becomes the first Health Canada-approved dual GIP (搜索)/GLP-1 (搜索) receptor agonist for chronic weight management in adults with obesity (搜索) or overweight conditions.
Clinical trial data from the SURMOUNT program demonstrated significant weight loss, with patients losing an average of 21.8 kg (48 lbs) at the highest 15 mg dose compared to 3.2 kg (7 lbs) on placebo.
The approval addresses a critical medical need in Canada where one in three adults live with obesity (搜索), a chronic disease with an estimated economic burden of $27.6 billion annually.
Eli Lilly Canada (搜索) announced that Zepbound KwikPen (tirzepatide injection) has received Health Canada authorization and is now available as the first dual-receptor agonist approved for obesity (搜索) treatment in the country. The once-weekly injection represents a significant advancement in chronic weight management, targeting both GIP (搜索) (glucose-dependent insulinotropic polypeptide) and GLP-1 (搜索) (glucagon-like peptide-1) hormone receptors.
Clinical Efficacy Demonstrated in Phase 3 Trials
The approval was supported by robust clinical evidence from the SURMOUNT phase 3 program, which enrolled more than 5,000 participants across six registration studies. In the pivotal SURMOUNT-1 trial involving 2,539 adults with obesity (搜索) or excess weight and weight-related medical conditions, tirzepatide demonstrated substantial weight reduction compared to placebo at 72 weeks.
Participants receiving the highest dose of 15 mg lost an average of 21.8 kg (48 lbs), while those on the 5 mg dose achieved an average weight loss of 15.4 kg (34 lbs). In contrast, the placebo group lost only 3.2 kg (7 lbs) on average. The treatment was administered as an adjunct to diet and exercise modifications.
Addressing Canada's Obesity Crisis
The approval comes at a critical time for Canadian healthcare, as obesity (搜索) affects one in three Canadians and represents a chronic, progressive disease resulting from complex genetic, environmental, behavioral, biological, and social factors. The condition is associated with more than 200 weight-related health conditions, including heart disease, type 2 diabetes (搜索), and various cancers.
"The evidence and research are unequivocal, obesity (搜索) is a chronic disease with severe health implications and when not properly addressed, leads to numerous other chronic conditions, strains health systems, reduces productivity, and increases disability," said Mathilde Merlet, General Manager and President of Lilly Canada.
The economic impact of untreated obesity (搜索) in Canada has reached $27.6 billion, representing a 20% increase from previous estimates. Beyond financial costs, patients face substantial bias and stigma, with 54% of adults with obesity reporting workplace stigmatization and 64% experiencing weight bias from healthcare professionals.
Treatment Indication and Mechanism
Health Canada authorized Zepbound KwikPen for chronic weight management in adults with obesity (搜索) (BMI ≥ 30 kg/m²) or overweight individuals (BMI 27-30 kg/m²) who have at least one weight-related comorbid condition such as hypertension (搜索), dyslipidemia (搜索), type 2 diabetes (搜索) mellitus, obstructive sleep apnea (搜索), or cardiovascular disease (搜索).
The drug's unique dual-receptor mechanism distinguishes it from existing GLP-1 (搜索)-only treatments. By activating both GIP (搜索) and GLP-1 receptors, tirzepatide provides enhanced metabolic effects for weight management, including both weight loss and weight maintenance phases.
Healthcare Professional and Advocacy Response
The medical community has welcomed the approval as expanding treatment options for obesity (搜索) management. Dr. Sean Wharton, an Internal Medicine Physician and Weight Management Specialist, noted that "This will increase the options for weight management. This is a win for Canadians who struggle with obesity and obesity related conditions."
Dr. Yves Robitaille, an Obesity (搜索) Management Specialist, emphasized the medication's role in achieving therapeutic targets: "Managing obesity often requires medication to reach targets that will improve people's health. Health Canada's approval of tirzepatide for the treatment of this disease allows us to add this tool to our options, thus providing Canadian patients with an effective new medicine to manage their obesity."
Patient advocacy organizations have also expressed support for the approval. Lisa Schaffer, Executive Director at Obesity (搜索) Canada, described the development as marking "a new era in obesity care — one defined by innovation, growing public awareness, and the willingness to challenge outdated thinking."
Market Access and Availability
Zepbound KwikPen received Health Canada authorization on May 13, 2025, and is now commercially available through various channels. The treatment joins the growing class of incretin-based therapies that have transformed metabolic disease management, though questions remain about accessibility and insurance coverage for this chronic condition treatment.
The SURMOUNT clinical development program, which began in late 2019, included four global registration studies (SURMOUNT-1, SURMOUNT-2, SURMOUNT-3, and SURMOUNT-4) that collectively demonstrated tirzepatide's significant weight reduction benefits compared to placebo across diverse patient populations with and without type 2 diabetes (搜索).
