Clinical Significance of MIPLYFFA
MIPLYFFA's approval marks a significant advancement in the treatment of NPC, a rare, progressive, neurodegenerative disorder characterized by the body's inability to transport cholesterol and other lipids within cells. This dysfunction leads to the accumulation of these substances, affecting various cell types, including neurons, and resulting in significant physical and cognitive impairments, often leading to early mortality. Mechanism of Action
MIPLYFFA functions by increasing transcription factors EB (TFEB) and E3 (TFE3) activation, leading to the upregulation of genes associated with lysosomal expression and regulation (CLEAR). Additionally, the medication has shown a reduction in unesterified cholesterol in lysosomes of human NPC fibroblasts, although the clinical significance of this finding is not fully understood.
