Neratinib Shows Anti-Atherosclerotic Effects Through Novel ASK1 Pathway in Preclinical Studies

- Neratinib, a breast cancer drug, demonstrated significant reduction in vascular inflammation and atherosclerosis in preclinical mouse models by targeting ASK1 rather than its traditional HER2 pathway. - The drug suppressed endothelial inflammation triggered by multiple stimuli and reduced plaque burden, necrotic core size, and macrophage infiltration in atherosclerotic lesions. - Combination therapy with neratinib and rosuvastatin produced superior anti-atherosclerotic effects compared to statin monotherapy, reducing multiple inflammation-related cytokines and chemokines. - These findings suggest potential for drug repurposing neratinib as a novel anti-inflammatory approach for cardiovascular disease, though human clinical trials are needed.
