RIG-101 is an intranasal stem-loop RNA (SLR) therapeutic that agonizes RIG-I, triggering type I and type III interferon production in the respiratory epithelium. RIG-101 is an oligonucleotide of the stem-loop RNA therapeutic (SLR) class that agonizes RIG-I, a cytosolic innate immune receptor. Activation of RIG-I triggers type I and type III interferon production in the respiratory epithelium, priming innate antiviral defenses at the mucosal surface. In asthma patients, intranasal RIG-101 induced type I and type III interferons and upregulated interferon-stimulated genes in the upper respiratory tract, a pharmacodynamic signature consistent with the intended mechanism of action.
