Methylglyoxal (MG) directly activates GABA-A receptors on neurons, producing a rapid inhibitory effect. This represents a previously unknown mechanism linking neuronal inhibitory tone to metabolic activity. MG is an endogenous byproduct of glycolysis that is normally metabolized by glyoxylase 1 (Glo1); inhibiting Glo1 raises MG levels, which in turn enhances GABA-A receptor-mediated inhibition.
