89bio Advances Phase 3 Trials for Pegozafermin in MASH and Severe Hypertriglyceridemia with $561M Cash Position
核心洞察
89bio reported Q2 2025 financial results with $561.2 million in cash, cash equivalents, and marketable securities as of June 30, 2025.
The company expects topline histology data from ENLIGHTEN-Fibrosis Phase 3 trial in non-cirrhotic MASH (搜索) patients in first half of 2027 and ENLIGHTEN-Cirrhosis data in 2028.
ENTRUST Phase 3 trial topline data for pegozafermin in severe hypertriglyceridemia (搜索) is anticipated in first quarter of 2026.
89bio, Inc. (Nasdaq: ETNB) reported second quarter 2025 financial results and provided updates on its Phase 3 clinical development program for pegozafermin, a fibroblast growth factor 21 (FGF21 (搜索)) analog being evaluated for metabolic dysfunction-associated steatohepatitis (搜索) (MASH (搜索)) and severe hypertriglyceridemia (搜索) (SHTG (搜索)). The San Francisco-based clinical-stage biopharmaceutical company maintained a strong cash position of $561.2 million as of June 30, 2025.
"As we enter the second half of 2025, we remain diligently focused on execution across our three global Phase 3 trials for pegozafermin in MASH (搜索) and SHTG (搜索)," said Rohan Palekar, Chief Executive Officer of 89bio. "As our clinical development program in MASH continues to progress, we believe pegozafermin is poised to potentially deliver best-in-class relative risk ratio on histology endpoints, a favorable safety and tolerability profile, and a convenient dosing approach to the large and underserved MASH patient population."
Phase 3 MASH Program Progress
The company's two Phase 3 MASH (搜索) trials continue patient enrollment with key data readouts expected over the next three years. The ENLIGHTEN-Fibrosis trial in non-cirrhotic (F2-F3) MASH patients is expected to deliver topline histology data in the first half of 2027, while the ENLIGHTEN-Cirrhosis trial in compensated cirrhotic (F4) MASH patients will report results in 2028.
89bio anticipates that data from the histology cohorts of both ENLIGHTEN trials will support accelerated approval applications for pegozafermin in both non-cirrhotic (F2-F3) and compensated cirrhotic (F4) MASH (搜索) patient populations. This regulatory strategy has been aligned with both the U.S. Food and Drug Administration (FDA) and European Medicines Agency (EMA). Following the histology readouts, both trials will continue to collect outcomes data to potentially support full approval.
Severe Hypertriglyceridemia Trial Design
The ENTRUST Phase 3 trial represents a randomized, double-blind, placebo-controlled global study evaluating pegozafermin's efficacy, safety, and tolerability in SHTG (搜索) patients. The trial randomizes patients to pegozafermin (30 mg or 20 mg) or placebo in a 3:3:2 ratio, administered once weekly for 52 weeks. The primary endpoint measures the percent change from baseline in fasting triglycerides at Week 26 compared to placebo, with topline data expected in the first quarter of 2026.
Financial Performance and Manufacturing Investment
89bio's second quarter 2025 research and development expenses totaled $103.9 million, representing a significant increase from $44.9 million in the same period of 2024. This increase was primarily driven by the ongoing Phase 3 ENLIGHTEN trials in MASH (搜索) and a non-recurring payment of $42.4 million related to the construction of a commercial-scale production facility for pegozafermin.
The commercial manufacturing facility construction remains on schedule to support the company's Biologics License Application (BLA) filing for pegozafermin. 89bio's remaining financial obligation for the facility is limited to a final milestone payment of $13.5 million, scheduled for payment in 2026 upon completion of construction.
General and administrative expenses increased to $11.9 million in Q2 2025 from $8.6 million in the prior year period, primarily due to increased personnel-related expenses including stock-based compensation driven by higher headcount.
The company reported a net loss of $111.5 million for the three months ended June 30, 2025, compared to a net loss of $48.0 million for the same period in 2024. This increased loss was primarily attributed to higher R&D expenses associated with contract manufacturing, advancement of the Phase 3 MASH (搜索) programs, and personnel-related expenses.
Pegozafermin's Therapeutic Profile
Pegozafermin is described as a specifically engineered, potentially best-in-class FGF21 (搜索) analog featuring unique glycoPEGylated technology that optimizes biological activity through an extended half-life. The drug candidate is being developed for patients with liver and cardiometabolic diseases who currently lack optimal treatment options, including those with MASH (搜索) with advanced fibrosis and patients with compensated cirrhosis.
