Independent Validation Confirms Platform Efficacy
The Victorian Centre for Functional Genomics (搜索) at the Peter MacCallum Cancer Centre (搜索) conducted a comprehensive validation study of AlgoraeOS v1, testing 21 AI-predicted drug combinations across four cancer (搜索) cell models. The program generated approximately 10,000 data points to assess predicted synergy, representing a rigorous real-world evaluation of the platform's capabilities.
The study assessed 84 cannabidiol–drug–cell combinations, including both predicted synergistic and non-synergistic pairs, using four established synergy models: Highest Single Agent, Bliss independence, Zero Interaction Potency and Loewe additivity. AlgoraeOS v1 demonstrated strong predictive alignment with three of the four independent models and successfully excluded most combinations identified as non-synergistic.
"The results mark an important milestone in the real-world testing of the AOS1 in silico predictions," said Chief Scientific Officer Dr. James McKenna. Several drug combinations met predefined thresholds for synergy during testing and will now be evaluated for further in vitro or in vivo assessment.
AlgoraeOS v2 Delivers Enhanced Performance
Parallel to the validation program, Algorae launched AlgoraeOS v2, a major upgrade developed in collaboration with the University of New South Wales and CSIRO Data61 (搜索). The enhanced platform has been trained on more than 5.5 million inhibition records and models full dose–response surfaces across all four synergy formalisms while providing confidence-weighted predictions.
From a dataset spanning more than 500,000 potential cannabidiol–drug–cell line combinations, AlgoraeOS v2 identified 90 high-quality drug-combination candidates. The platform evaluates cannabidiol in combination with more than 3,000 approved and investigational drugs across 170 cell lines, incorporating confidence-weighted metrics that quantify both data-driven and model-driven uncertainty.
Benchmarking results show AlgoraeOS v2 outperforming representative state-of-the-art models, including those developed by Google DeepMind (搜索), with lower error and stronger correlation across key prediction metrics. UNSW AI Institute Associate Professor Fatemeh Vafaee noted that the platform "offers sovereign, high-performance capability that outperforms global state-of-the-art models while remaining lightweight and deployable."
Strategic Advancement and Commercial Pipeline
The validation study provided critical insights that are being integrated into the continued evolution of AlgoraeOS through refinements to internal ranking functions, reference models, and threshold settings. The data will inform the design of future preclinical programs aimed at efficiently identifying high-value synergistic combinations.
Dr. McKenna emphasized that the combination of improved predictive capabilities and enhanced granularity provides "a powerful approach for prioritising candidates for further interrogation and progression to in vitro validation assays."
Algorae plans to further prioritize the 90 potential candidates by incorporating commercial and intellectual-property considerations before progressing selected combinations into preclinical validation assays. The company has commenced discussions with the Peter MacCallum Cancer Centre (搜索) regarding a second program and independent validation of AlgoraeOS v2 results.
The company expects to release the first in silico predictions from AlgoraeOS v2 in December 2025, which will guide the next phase of preclinical evaluation including identification of new synergistic drug combinations and prioritization of repurposed candidates. Algorae also continues to progress its commercial pharmaceutical business, with additional products and agreements moving through the pipeline.
