The clinical-stage biotechnology company presented data demonstrating the therapeutic potential of this novel immunotherapy approach for treating CEACAM5 (搜索)-positive cancers.
Dual-Target Mechanism Shows Strong Anti-Tumor Activity
ATOR-4066 is engineered as a Neo-X-Prime bispecific antibody that simultaneously targets CD40 (搜索) and CEACAM5 (搜索), designed to activate the immune system against CEACAM5-positive cancers such as colorectal and gastric cancer (搜索). The preclinical data presented in poster #253 revealed that ATOR-4066 strongly stimulates both myeloid cells and T cells inside tumors, resulting in powerful and durable anti-tumor effects.
According to the study results, ATOR-4066 eliminated established tumors in preclinical models while showing limited immune activation outside the tumor microenvironment. This targeted approach suggests the potential for reduced systemic toxicity while maintaining therapeutic efficacy.
Enhanced Efficacy with Combination Therapy
The preclinical findings demonstrated that ATOR-4066 showed enhanced efficacy when combined with PD-1 (搜索) blockade therapy. This synergistic effect supports the potential for combination treatment strategies that could improve patient outcomes in solid tumor malignancies.

ATOR-4066 represents an advancement in Alligator's CD40 (搜索)-focused therapeutic platform, building on the clinical progress achieved with mitazalimab, the company's lead drug candidate. Mitazalimab is currently ready for Phase 3 development and has previously demonstrated unprecedented survival data at 24-months follow-up in first-line metastatic pancreatic cancer (搜索) patients in the Phase 2 OPTIMIZE-1 trial.
