Almirall Launches Phase III Trial Testing Lebrikizumab for Nummular Eczema Treatment
核心洞察
Almirall announces the LumiNE Phase III clinical trial to evaluate lebrikizumab's efficacy in treating nummular eczema (搜索), a debilitating chronic skin condition distinct from atopic dermatitis (搜索).
The study will enroll at least 270 patients across 60 European centers for up to 48 weeks, targeting the IL-13 (搜索) cytokine pathway that plays a central role in nummular eczema (搜索) pathogenesis.
Nummular eczema (搜索) affects 0.1% to 9% of the population with coin-shaped, itchy patches that significantly impair quality of life and are often difficult to manage with standard topical therapies.
Almirall has announced the initiation of LumiNE, a Phase III clinical trial evaluating lebrikizumab (Ebglyss®) for the treatment of nummular eczema (搜索), marking a significant expansion of the drug's potential therapeutic applications beyond its current approval for moderate-to-severe atopic dermatitis (搜索). The study will begin in the first half of 2026 and represents part of Almirall's ongoing lifecycle management activities to extend advanced treatments to patients with dermatological conditions characterized by high unmet medical needs.
Study Design and Patient Population
The LumiNE trial (M-27501-30) is designed as a randomized, double-blind, placebo-controlled, parallel group, multicenter clinical trial with a double-blind extension. The study will evaluate the efficacy, safety, and tolerability of lebrikizumab for up to 48 weeks of treatment in adult participants with nummular eczema (搜索) who are not adequately controlled with topical corticosteroids or when such treatment is not medically advisable.
The trial will initially randomize approximately 270 participants at around 60 centers across Europe, with potential for extension to additional patients. The primary endpoint focuses on improvement of IGA-NE (Investigator's Global Assessment of nummular eczema (搜索)), while key secondary endpoints include pruritus NRS (Numerical Rating Scale) improvement from baseline and absolute change in DLQI (Dermatology Life Quality Index).
Scientific Rationale for IL-13 Targeting
Nummular eczema (搜索), while distinct from atopic dermatitis (搜索), involves similar disease mechanisms with the cytokine IL-13 (搜索) playing a key role. Recent scientific data demonstrate that IL-13 serves as a central mediator in nummular eczema, playing an important role in disease onset and relapse by specifically contributing to triggering inflammation, pruritus, eosinophilic infiltration, and barrier dysfunction.
"The proven efficacy of lebrikizumab in atopic dermatitis (搜索), its high affinity to the cytokine IL-13 (搜索), and the scientific evidence pointing to a central role of IL-13 also in nummular eczema (搜索) could enable us to expand the benefits of lebrikizumab to these patients," stated Karl Ziegelbauer, Chief Scientific Officer at Almirall.
Disease Burden and Clinical Need
Nummular eczema (搜索) is a chronic skin disorder characterized by red, scaly, itchy, coin-shaped patches that are often difficult to manage with standard topical therapies. The condition, also referred to as discoid eczema, may present in individuals with or without atopic dermatitis (搜索) and represents a frequently relapsing inflammatory skin condition that significantly impairs patients' quality of life due to persistent itch, sleep disruption, and social stigma.
The disease has an estimated prevalence range from 0.1% to 9%, with higher prevalence observed in older adults and specific ethnic populations. Nummular eczema (搜索) is frequently misdiagnosed due to its resemblance to other inflammatory dermatoses, and a growing body of evidence suggests it requires specific tailored diagnostic and therapeutic approaches.
Lebrikizumab Mechanism and Clinical Profile
Lebrikizumab is a high-affinity monoclonal antibody that selectively binds the cytokine IL-13 (搜索), preventing the formation of the IL-13Rα1/IL-4Rα (搜索) heterodimer complex and subsequent signaling. This mechanism inhibits the biological effects of IL-13 and its role in disease pathogenesis. The drug represents a step forward for patients not controlled with topical therapy due to its selective mechanism of action, proven efficacy, favorable safety profile, and 4-weekly maintenance dosing regimen.
Lebrikizumab has demonstrated clinical efficacy in both monotherapy and in combination with topical corticosteroids across various clinical studies and real-world evidence settings. Ebglyss was approved in Europe in October 2023 and in the US in August 2024 for moderate-to-severe atopic dermatitis (搜索) treatment.
Clinical Leadership Perspective
Prof. Dr. Kilian Eyerich, Medical Director of the Department of Dermatology and Venereology at the Medical Center University of Freiburg, Germany, and LumiNE coordinating investigator, emphasized the study's importance: "This is an important clinical study as it is aimed at addressing the unmet needs of patients living with nummular eczema (搜索), and the latest scientific insights that show a potential role of IL-13 (搜索) in the development of this debilitating chronic skin disease."
The trial aligns with Almirall's strategic focus on medical dermatology leadership and represents the company's commitment to transforming patients' lives through innovative therapeutic approaches targeting conditions with significant unmet medical needs.
