Revolutionary Manufacturing Platform Gains FDA Recognition
The expansion centers on Alnylam's next-generation siRELIS™ (siRNA Enzymatic Ligation Synthesis) platform, which has been accepted into the U.S. Food and Drug Administration's Emerging Technology Program. This acceptance accelerates dialogue with global health authorities on innovative manufacturing approaches for oligonucleotide-based medicines and follows successful demonstration through production of pilot-scale batches of zilebesiran and nucresiran.
Timothy Maines, Chief Technical Operations and Quality Officer at Alnylam, explained the transformative impact: "We're going to go from being able to make kilogram quantities [of medicines] to metric-ton quantities — that's a game changer, right? It will also help us with some of the cost of making the medicines."
The siRELIS™ platform represents a breakthrough in manufacturing efficiency, assembling short, high-quality RNA fragments called "blockmers" into complete molecules more efficiently than traditional methods. This scalable approach reduces use of starting materials and reliance on organic solvents while increasing capacity in a single facility.
Targeting Common Diseases with Expanded Capacity
While Alnylam currently markets drugs treating rare kidney, heart and liver disorders, along with a recently FDA-approved hemophilia (搜索) treatment, the company is developing therapeutics for larger patient populations. The expanded manufacturing capabilities will support pipeline programs targeting hypertension (搜索), fatty liver disease (搜索), diabetes, and obesity (搜索).

Facility Expansion Details and Economic Impact
The $250 million expansion of the 200,000 square foot Norton facility will increase capabilities to produce both clinical and commercial supply of siRNA oligonucleotide drug substance for global distribution. The facility, which opened in 2021, currently employs approximately 125 people and serves as a pipeline accelerator for early-stage programs targeting different tissue types including liver, CNS, muscle, and adipose tissue.
Construction is underway with new capabilities expected to become fully operational by late 2027. The expansion will add about one-third more active manufacturing space and is expected to create new positions in manufacturing, facilities, engineering, finance, and IT support.

The company employs roughly 2,500 people globally, with about 1,200 working in Massachusetts across its Kendall Square headquarters, Cambridge Alewife facility, and Norton manufacturing plant.
