Targeting Difficult-to-Drug Proteins in Acute Leukemia
The acquisition centers on Dark Blue's lead program DBT-3757 (搜索), a potentially first-in-class small molecule designed to target MLLT1 (搜索) and MLLT3 (搜索), transcriptional regulators linked to abnormal gene expression in acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL). Rather than inhibiting these proteins through traditional approaches, Dark Blue employs targeted protein degradation to remove the proteins from cells entirely.
"The targets MLLT1 (搜索) and MLLT3 (搜索) are well-studied in academic research, but translating that biology into drugs has been challenging," according to the acquisition details. This strategy is particularly relevant for targets that have proven resistant to classical small molecule inhibition, potentially expanding what's addressable in leukemia treatment.
Addressing Critical Unmet Medical Need
The acquisition targets a significant clinical challenge in acute leukemia, where relapse remains common despite initial treatment responses. In AML, relapse after chemotherapy occurs in over 50% of younger patients and up to 90% of elderly patients. Outcomes for relapsed/refractory disease remain poor, with reported five-year survival around 10%.
"Acute myeloid leukaemia remains one of the most difficult cancers to treat, and we see an urgent need for new mechanisms capable of changing the trajectory of this disease," said Jay Bradner, Amgen's head of R&D.
Strategic Early-Stage Investment
Amgen framed the acquisition as part of its strategy to strengthen internal oncology discovery by adding new biology and expertise at an early stage, rather than acquiring late-stage clinical assets. The company is stepping in at a preclinical stage, willing to absorb early discovery risk in exchange for greater control over how the biology is developed.
Beyond the lead program, Dark Blue has been developing other small molecules, including projects against targets such as ADAR1 (搜索), an enzyme commonly overexpressed in cancer, and SMO (搜索), a protein in the Hedgehog signaling pathway established as a target in diseases like AML and basal cell carcinoma.
Company Background and Integration Plans
Dark Blue Therapeutics (搜索) was formed in 2020 and emerged with a £75 million ($101 million) Series A in 2022 led by Forbion and backed by Oxford Science Enterprises, Bristol Myers Squibb (搜索), and Evotec. In March 2025, the company appointed serial biotech entrepreneur Edwin Moses as chairman.
"We experienced considerable interest from pharmaceutical companies in the lead programme," said Moses, who noted that the board had evaluated the relative merits of acquisition versus traditional venture capital financing. "We were delighted to reach agreement on the acquisition of the company by Amgen, who we are sure will be able to accelerate the development and broaden the applications of this lead programme."
Amgen plans to integrate Dark Blue into its existing R&D operations, which include hubs in Cambridge and Uxbridge in the UK. The immediate focus will be on advancing programs through IND-enabling work and into clinical development.
Broader Industry Context
The deal reflects continued large pharma interest in next-generation targeted approaches in oncology, including protein degradation. Recent examples include Gilead's partnership with Kymera (搜索), an option-based deal worth up to $750 million around molecular glue degraders in cancer.
Dark Blue's pipeline spans degraders, molecular glues, and inhibitors, with DBT-3757 (搜索) having potential as both monotherapy and in combination with other therapies for AML and solid tumors. The acquisition complements and extends Amgen's research in targeted protein degradation and leukemia therapeutics, following previous licensing deals with Plexium and Arrakis Therapeutics for protein degrader therapeutics.
