Mice treated with the combination of AOH1996 (100 mg/kg, BID, PO) and VEN (100 mg/kg, daily, PO) showed a significant increase in median survival (MS) compared to control, VEN alone, or AOH1996 alone. The median survival was 54 days for the combination group, compared to 34 days for the control and VEN groups, and 43.5 days for the AOH1996 group. Secondary transplants further confirmed the benefit of the combination, with an MS of 49.5 days for the combination group compared to 28, 32.5, and 41 days for the control, VEN, and AOH1996 groups, respectively.
Activity in FLT3-WT PDX AML Model
To assess the broader applicability of the AOH1996 and VEN combination, the researchers also tested its efficacy in a FLT3 (搜索)-WT PDX AML (搜索) model. Similar to the MllPTD/WT/Flt3ITD/ITD model, the combination of AOH1996 and VEN significantly prolonged survival in both primary and secondary transplanted leukemic mice. The median survival in primary recipients was 75.5 days for the combination group, compared to 41, 48, and 55 days for the control, VEN, and AOH1996 groups, respectively. In secondary recipients, the median survival was 76 days for the combination group, compared to 40, 51, and 60 days for the control, VEN, and AOH1996 groups, respectively.
These findings suggest that AOH1996, when combined with venetoclax, represents a promising therapeutic strategy for targeting leukemia (搜索) stem cells and improving outcomes in acute myeloid leukemia, including FLT3 (搜索)-WT AML (搜索).
