Dual-Targeting Rationale

In June 2026, the company was awarded a $1.5 million non-dilutive research grant from the Andy Hill Cancer Research Endowment (CARE) Fund to support IND-enabling work for APVO451 (搜索), a novel trispecific antibody-like immunotherapy candidate targeting nectin-4 for solid tumors.
APVO451 (搜索) brings together three functions in a single molecule: targeting nectin-4 (搜索), activating CD40 (搜索) on antigen-presenting cells to engage innate immunity, and engaging CD3 (搜索) on T-cells to direct tumor-cell killing. The trispecific candidate is designed so that its CD3 and CD40 activity depends on binding to nectin-4, helping focus immune activation within the tumor microenvironment and reduce the risk of systemic immune activation.
"By combining nectin-4 (搜索)-targeting with CD40 (搜索) and CD3 (搜索) engagement, APVO451 (搜索) is designed to activate both antigen-presenting cells and T cells within the tumor microenvironment," said Janatpour. APVO451 is advancing toward development candidate selection by year-end 2026, with IND-enabling studies anticipated to begin in the first quarter of 2027.
