The facility, combined with existing cash resources, extends Aptevo's funding runway into 2029.
Financial Structure and Strategic Benefits
The ELOC provides Aptevo with discretionary access to capital under market-based conditions, offering affordable financing with minimal fees and no warrants. "This agreement offers added flexibility and control over how and when we access capital," said Daphne Taylor, Chief Financial Officer at Aptevo. "Importantly, the fully leveraged facility plus cash on hand is sufficient to fund us for three years, into 2029. During this time, we plan to continue generating clinical data and advancing differentiated assets across the portfolio focusing on execution and value creation."
Under the agreement terms, Aptevo has the right, but not the obligation, to sell shares to the counterparty from time to time, subject to customary conditions and limitations. The company retains full discretion over facility usage and is not required to draw on the funding.
Lead Asset Shows Clinical Promise
Aptevo's portfolio is anchored by mipletamig, a first-in-class CD123 (搜索) x CD3 (搜索) bispecific currently being evaluated in RAINIER, a Phase 1b/2 trial for frontline acute myeloid leukemia (AML). The drug candidate has been evaluated in more than 100 patients across three trials, consistently demonstrating high remission rates and a favorable safety and tolerability profile, with no observed events of cytokine release syndrome in frontline patients treated to date.
Mipletamig is being studied in combination with standard-of-care venetoclax plus azacitidine for the treatment of frontline AML and has received orphan drug designation for AML according to the Orphan Drug Act.
Expanding Multispecific Platform
Building on clinical validation from mipletamig, Aptevo has developed a portfolio of tumor-directed CD3 (搜索) programs utilizing its proprietary ADAPTIR and ADAPTIR-FLEX platform technologies. The pipeline includes bispecific candidates APVO442 (PSMA (搜索) × CD3) for prostate cancer and APVO455 (搜索) (Nectin-4 (搜索) × CD3) for solid tumors, as well as trispecific candidates APVO451 (搜索) and APVO452 (搜索).
All candidates incorporate the company's proprietary application of the CRIS-7-derived CD3 (搜索) binding domain, designed to enable tumor-focused immune activation with potential for meaningful antitumor activity while prioritizing safety and tolerability.
Additional Pipeline Assets
The company maintains two clinical candidates beyond mipletamig. ALG.APV-527, a bispecific conditional 4-1BB (搜索) agonist designed to be active only upon simultaneous binding to 4-1BB and 5T4 (搜索), is being co-developed with Alligator Bioscience (搜索). The candidate was most recently evaluated in a Phase 1 clinical trial for multiple solid tumor types likely to express 5T4.
Aptevo also has six preclinical candidates with different mechanisms of action designed to target a range of solid tumors, all created from the company's two proprietary platforms.
