ASAP and Michael J. Fox Foundation Announce $261M Investment to Advance Personalized Parkinson's Treatments
核心洞察
Aligning Science Across Parkinson's (搜索) (ASAP) and The Michael J. Fox Foundation (搜索) have launched a $261 million global research initiative to develop personalized treatments for Parkinson's disease (搜索).
The expanded Collaborative Research Network now includes 32 new research teams, bringing the total to 67 teams with nearly 400 investigators across 187 institutions in 24 countries.
Research teams will focus on understanding Parkinson's disease (搜索) heterogeneity, including environmental risk factors, co-pathologies, and the role of alpha-synuclein (搜索) in disease progression.
Aligning Science Across Parkinson's (搜索) (ASAP) and The Michael J. Fox Foundation (搜索) have announced a landmark $261 million investment to expand their global research initiative aimed at developing personalized treatments for Parkinson's disease (搜索). This significant funding round represents a major commitment to understanding the complex heterogeneity of Parkinson's disease and accelerating the development of targeted therapies.
Expanding Global Research Network
The investment will support 32 new research teams as part of ASAP's Collaborative Research Network (CRN) expansion. With this addition, ASAP has now funded a cumulative total of 67 CRN teams since its inception, creating an expansive network that encompasses nearly 400 investigators across 187 institutions in 24 countries.
"Solving a disease as complex as Parkinson's requires big bets on bold biological hypotheses," said Sonya Dumanis, PhD, managing director of ASAP. "We are at an inflection point where new capabilities, from high-resolution imaging to advanced computational modeling, allow us to pose more complex questions about factors like aging and the environment and how they drive the disease."
Each team will receive between $6 million and $9 million over three years, depending on the research theme. This multi-year structure empowers collaborative groups of three to five investigators to combine their expertise and tackle high-priority research questions to shorten the timeline from laboratory insight to clinical application.
Targeting Disease Heterogeneity
The new research teams will focus on exploring the heterogeneity of Parkinson's disease (搜索), including when it starts, what symptoms appear, and how quickly it progresses. The research initiatives will address several critical areas:
Environmental Risk Mapping
Teams will study how exposure to pesticides and air pollution interact with genetics to cause and accelerate Parkinson's disease (搜索), using tools like AI-driven molecular fingerprinting.
Co-pathology Investigation
Researchers will investigate how multiple protein abnormalities commonly associated with neurodegenerative diseases such as Alzheimer's (搜索) and ALS (搜索), including tau (搜索), amyloid-beta (搜索), and TDP-43, interact with Parkinson's pathology to drive disease in individual patients.
Aging and Immune System Changes
Teams will explore aging-related changes to the immune system and genes, and their role in vulnerability to Parkinson's disease (搜索).
Brain Circuit Analysis
Research will reveal how brain circuits disrupted by pathological alpha-synuclein (搜索) contribute to symptoms such as sleep disorders and cognitive dysfunction.
Alpha-synuclein Seeding Research
Teams will characterize the seeding agent driving the alpha-synuclein (搜索) seeding amplification assay (SAA) — a transformative diagnostic for Parkinson's disease (搜索). By doing so, researchers aim to decode specific disease subtypes and illuminate the systemic biological mechanisms of the disease as it manifests throughout the body.
Brain Clearance Mechanisms
Research will study the brain's clearance mechanisms to define how these mechanisms' breakdown contributes to Parkinson's and uncover new therapeutics.
Tool Development for Research Community
Several new teams will focus on tool generation, developing novel resources for the research community — including CRISPR-engineered cell lines, viral vectors, and advanced chemical probes — to support continued work on emerging targets identified in previous ASAP awards. By providing these standardized, validated resources to the global community, ASAP is helping remove technical barriers that can slow early-stage research and drug development.
Urgent Need for Better Treatments
"For people living with Parkinson's, the need for better treatments has never been more urgent," said Todd Sherer, PhD, chief mission officer at The Michael J. Fox Foundation (搜索). "This level of coordinated, global investment brings together leading scientists to tackle the most critical questions about the disease; not in isolation, but as a connected community working to accelerate discovery and move new ideas into the treatment pipeline."
The CRN expansion is designed to funnel innovative ideas into the research and development pipeline while systematically dismantling technical roadblocks. By accounting for the inherent heterogeneity of the disease to validate targets within a human biological context, the initiative aims to accelerate the development of the next generation of personalized therapies.
Foundation's Broader Impact
As the world's largest nonprofit funder of Parkinson's disease (搜索) research, The Michael J. Fox Foundation (搜索) has funded $3 billion in research to date and fundamentally altered the trajectory of progress toward a cure. The Foundation operates at the hub of worldwide Parkinson's research, forging groundbreaking collaborations with industry leaders, academic scientists and government research funders.
ASAP is an initiative of the CNS Quest, a program of the Sergey Brin Family Foundation (搜索) and Catalyst4 to accelerate discoveries and improve daily life for people impacted by Parkinson's disease (搜索), bipolar disorder, and autism. ASAP is managed by the Coalition for Aligning Science (搜索) and partners with The Michael J. Fox Foundation (搜索) for Parkinson's Research to implement its programs.
