Enhanced Production Capabilities
The two enhancers were developed through collaborative research efforts and tested at multiple scales in HEK293 cells using both Ascend's EpyQ® two-plasmid AAV (搜索) Production System and a commercially available 3-plasmid system. Notably, no negative effects on quality parameters such as vg/cap ratio or mispackaged DNA were observed during testing.
Enhancer 1 demonstrated remarkable performance improvements, producing up to 3-fold more AAV (搜索) vector using EpyQ® in Ambr®15 systems and up to 1.7-fold more for the 3-plasmid system at Ambr®15 scale and in 2L bioreactors. Enhancer 2 showed similarly impressive results, producing up to 2.4-fold more AAV using the EpyQ® plasmid system at Ambr®15 scale, and up to 2.1-fold increases for Ambr®250 scale and 5L bioreactors.
The enhancer activity was further validated using customer plasmids, and the work has been replicated in experiments conducted by an external party interested in licensing the technology.
