Aspen Neuroscience's Personalized Parkinson's Cell Therapy Shows Promise in Phase II Trial, Advances to Pivotal Study
核心洞察
Aspen Neuroscience (搜索)'s autologous stem cell therapy sasineprocel (搜索) demonstrated improved motor function and quality of life in eight Parkinson's disease (搜索) patients after 12 months of treatment.
The therapy showed mean improvements in "ON" time of 2.1-2.4 hours and significant reductions in MDS-UPDRS motor scores, with no serious surgical adverse events observed.
Based on these encouraging Phase Ia/II results, the company plans to advance sasineprocel (搜索) to a Phase III pivotal trial later this year.
Aspen Neuroscience (搜索)'s personalized stem cell therapy for Parkinson's disease (搜索) has demonstrated encouraging clinical benefits in early-stage trials, positioning the company to advance its lead candidate sasineprocel (搜索) into pivotal Phase III testing later this year.
Data from the first eight patients in the Phase Ia/II ASPIRO trial (NCT06344026) showed meaningful improvements in motor function and quality of life measures after 12 months of treatment. The results were presented at the AD/PD 2026 International Conference on Alzheimer's and Parkinson's diseases in Copenhagen, Denmark.
Clinical Outcomes Show Motor Function Improvements
The trial evaluated sasineprocel (搜索), an autologous induced-pluripotent stem cell (iPSC)-derived dopaminergic neuron precursor cell therapy, in two dosing cohorts of four patients each. Both low-dose and high-dose groups demonstrated numerical improvements across multiple clinical endpoints.
Patients experienced a mean increase in "Good ON" time of 2.1 hours in the low-dose cohort and 2.4 hours in the high-dose group. The "ON" time refers to periods when Parkinson's medications are working effectively and symptoms are well-controlled.
Motor function assessments using the MDS-UPDRS III scale showed substantial improvements, with OFF scores reducing by 15.5 points in the low-dose cohort and 13.5 points in the high-dose group. The MDS-UPDRS II scores, which measure activities of daily living, improved by 5.3 points at low dose and 2.3 points at high dose.
Quality of life measures using the PDQ-39 scale demonstrated mean improvements of 51.6% in the low-dose group and 28.5% in the high-dose cohort.
Safety Profile Supports Continued Development
The therapy demonstrated a favorable safety profile with no serious surgical adverse events reported. Notably, no cases of severe graft-induced dyskinesia were observed, addressing a key concern with cell replacement therapies. Additionally, no symptomatic hemorrhages or infarctions occurred during the treatment period.
"We're very encouraged by these data showing improvements in function, physician- and patient-reported outcomes, and quality-of-life, and look forward to advancing sasineprocel (搜索) to a Phase III study later this year," said Dr. Revati Shreeniwas, Aspen's Chief Medical Officer.
Personalized Approach Differentiates Technology
Aspen's therapeutic approach involves converting patients' own skin cells into stem cells, which are then differentiated into implantable dopamine-producing cells designed to replace damaged neurons in the brain. The cells are precisely delivered to the putamen (搜索), a brain region critical for motor control that is significantly affected in Parkinson's disease (搜索).
"These findings support our approach of reprogramming a patient's own cells to wind back the epigenetic clock to a pre-disease state," Dr. Shreeniwas explained. "Combined with precisely delivering our multi-cell composition to the putamen (搜索), we're starting to see the positive clinical impact of rebuilding the neural networks damaged by this debilitating disease."
Competitive Landscape in Parkinson's Cell Therapy
The Parkinson's cell therapy space is becoming increasingly competitive, with multiple companies advancing different approaches. BlueRock Therapeutics (搜索), owned by Bayer (搜索), is investigating bemdaneprocel, an off-the-shelf cell therapy using donor cells that is currently in Phase III trials. While this approach enables easier mass manufacturing, it requires patients to undergo immunosuppressive treatments.
Hope Biosciences Research Foundation (搜索) is also preparing to advance its Parkinson's cell therapy into Phase III trials following positive Phase II results. Additionally, Kenai (搜索) recently initiated dosing in a Phase I trial of its cellular neuron replacement therapy RNDP-001 for moderate-to-severe idiopathic Parkinson's disease (搜索), with initial safety and brain imaging data expected this year.
Market Outlook and Unmet Need
According to GlobalData forecasts, the Parkinson's disease (搜索) market is projected to reach $7.9 billion across seven major markets by 2033. David Dexter, head of research at Parkinson's UK, noted that while researchers and drugmakers are making progress in characterizing therapies for Parkinson's, multiple factors still need to be addressed to facilitate continued advancement in the field.
