Novel STING Pathway Approach
NanoSTING-001 represents a new class of host-directed therapeutics designed to activate the innate immune response within the respiratory tract. The treatment uses lipid nanoparticles to deliver a natural activator of the stimulator of interferon genes (STING (搜索)) pathway, providing loco-regional immune enhancement. This approach is intended for use both before and after pathogen exposure to prevent respiratory infections and reduce severe illness risk.
The therapeutic is being developed to provide broad-spectrum protection independent of the specific respiratory pathogen, offering potential advantages over pathogen-specific treatments. Host-directed therapies targeting the innate immune system achieve protection independent of vaccination and hold promise for prevention and treatment of illnesses from multiple respiratory threats.
Preclinical Efficacy Data
Preclinical testing has demonstrated encouraging results for NanoSTING-001's safety and efficacy profile. The candidate therapeutic appears well tolerated, rapidly activates innate immunity, and treats consequences of infection. Notably, intranasal NanoSTING-001 has shown effectiveness against influenza A and B, including strains resistant to Oseltamivir (Tamiflu) and highly pathogenic avian influenza H5N1.
The upcoming swine studies will assess NanoSTING-001's ability to protect pigs from influenza infection, providing crucial efficacy data to inform clinical trial design. These validated animal models represent an important step toward human testing of the therapeutic.
Addressing Respiratory Disease Burden
The development comes as respiratory pathogens continue to impose significant clinical and economic burdens. During the 2024-2025 season in the United States, the CDC estimates 47 to 82 million flu illnesses, 610,000 to 1.3 million influenza-related hospitalizations, and 27,000 to 130,000 influenza deaths.
