Significant Reduction in Skin Disease Activity
The study demonstrated consistent and statistically significant reductions in cutaneous disease activity using validated dermatologic scoring systems. Mean CLASI scores decreased from 8.76 at baseline to approximately 5.6 at three months and fell below 4 by 12 months. These improvements were largely maintained through extended follow-up periods, including assessments at 36, 48, and 60 months.
Clinical response rates reinforced these findings across multiple timepoints. A CLASI-20 response, defined as at least 20% improvement in skin activity, was observed in most patients by six months and persisted throughout follow-up. Skin biopsies from four patients provided molecular insights into belimumab's mechanism of action. All patients received belimumab for a minimum of six months, either subcutaneously or intravenously, with follow-up extending up to five years. Cutaneous disease activity was assessed using both the CLASI and Revised CLASI scoring systems at baseline, 3- and 6-month intervals during the first year, and annually thereafter.
