Bloom Science Initiates Phase 1b Obesity Trial of BL-001, Advancing Ketogenic-Based Alternative to GLP-1 Therapies
核心洞察
Bloom Science (搜索) has dosed the first patient in a Phase 1b randomized, placebo-controlled trial evaluating BL-001, an oral Live Biotherapeutic Product designed to replicate ketogenic diet effects for obesity treatment.
The 12-week study will enroll up to 48 participants across two Australian clinical sites, targeting the majority of over 100 million American adults with obesity who remain untreated by current therapies.
Market research among thousands of U.S. adults shows substantial interest in ketogenic-metabolic approaches, with respondents preferring the BL-001 concept over GLP-1-based treatments due to its oral delivery and gradual weight loss profile.
Bloom Science (搜索) has initiated dosing in a Phase 1b clinical trial evaluating BL-001, an investigational oral Live Biotherapeutic Product designed to replicate the metabolic effects of the ketogenic diet for obesity treatment. The San Diego-based clinical-stage biotech company announced that the first patient has been dosed in the randomized, double-blind, placebo-controlled study.
Novel Approach Targets Untreated Obesity Population
The Phase 1b study (BL-001-1201) will evaluate the safety, tolerability, and weight loss effects of daily oral BL-001 administered for 12 weeks in adults with obesity. The trial will enroll up to 48 participants across two clinical sites in Australia, positioning BL-001 as a potential alternative to existing obesity treatments.
BL-001 represents a differentiated approach to obesity management, designed to deliver the therapeutic benefits of the ketogenic diet without requiring dietary restrictions. The therapy targets a significant unmet medical need, as the majority of more than 100 million American adults living with obesity remain untreated despite the availability of GLP-1 receptor agonists.
"Millions of patients remain untreated because existing therapies do not align with their preferences," said Christopher Reyes, PhD, Chief Executive Officer of Bloom Science (搜索). "BL-001 is designed to change that, delivering the metabolic benefits of the ketogenic diet in a daily oral capsule, without injections or dietary restriction."
Promising Early Clinical Data
Previous Phase 1 data in healthy adults demonstrated BL-001's tolerability profile, with no serious adverse events reported. Among overweight participants, the therapy achieved statistically significant placebo-adjusted weight loss, accompanied by dose-dependent BL-001 strain kinetics and metabolic changes consistent with engagement of ketogenic pathways.
The ketogenic diet, originally developed in 1921 to mimic fasting as an epilepsy treatment, has accumulated clinical evidence supporting its effectiveness for weight loss and reversal of metabolic dysfunction. However, the diet's long-term maintenance challenges have limited its broader therapeutic application.
Market Research Supports Differentiated Positioning
Recent market research conducted among several thousand U.S. adults, including a large cohort of individuals living with obesity, revealed substantial interest in ketogenic-metabolic approaches. This preference remained high even among those with prior GLP-1 experience, suggesting demand for therapeutic alternatives beyond current incretin-based treatments.
When presented with the BL-001 concept alongside GLP-1 profiles, respondents chose the BL-001 concept at higher rates than GLP-1-based approaches, with particular interest in the ketogenic mechanism. Respondents expressed clear preference for gradual, sustained weight loss with durable results over rapid initial weight loss associated with higher tolerability burden.
"This is about bringing a real treatment option to patients who are currently on the sidelines," said Louis Licamele, PhD, Chief Development Officer of Bloom Science (搜索). "Advancing into a dedicated obesity program is a milestone not just for Bloom, but for the patients who have been waiting for something different."
Broader Therapeutic Applications
BL-001 is being developed across both metabolic and neurologic indications, including obesity and Developmental & Epileptic Encephalopathies (DEEs). Preclinical data have demonstrated replication of the ketogenic diet therapeutic effect in epilepsy models and modulation of gamma-aminobutyric acid (GABA) and other key bioenergetic pathways to suppress hyperexcitability, a key driver in seizures.
The company's therapeutic approach is rooted in gut-brain axis biology and microbiome science, reflecting Bloom's strategy to leverage ketogenic biology across chronic diseases. BL-001 is positioned as a potential first-in-class, orally delivered Live Biotherapeutic Product candidate designed to translate ketogenic diet effects into a scalable, pharmaceutical-grade oral therapy.
Bloom Science (搜索) believes BL-001 has the potential to serve as stand-alone oral therapy while expanding the obesity market beyond patients currently treated with incretin therapies and offering complementary use alongside or following those treatments.
