Novel EGFR Inhibitor Shows Promise in Resistant Lung Cancer
BlossomHill's lead program, BH-30643, represents a first-in-class, orally available, macrocyclic, CNS-active, mutant-selective OMNI-EGFR (搜索) inhibitor designed for super-potency against a broad spectrum of EGFR kinase domain mutations. The drug targets EGFR classical mutations, atypical mutations such as PACC mutations, and diverse resistant mutations including C797S +/- T790M.
The company has commenced enrollment in expansion cohorts of the Phase 1/2 SOLARA trial, evaluating BH-30643 in both targeted therapy-naive and pretreated patients with locally advanced or metastatic EGFR (搜索)-mutant NSCLC. Preliminary data from the dose escalation portion, presented at the 2025 AACR-NCI-EORTC International Conference on Molecular Targets and Cancer Therapeutics, demonstrated systemic and CNS anti-tumor activity across various advanced EGFR-mutant NSCLCs with complex and difficult-to-treat resistance mutations.

BlossomHill has commenced combination enrollment with venetoclax in the first-in-human, Phase 1/1b dose escalation study of BH-30236 in relapsed or refractory acute myeloid leukemia (R/R AML) and higher-risk myelodysplastic syndrome (HR-MDS). A trial-in-progress poster was recently presented at the 67th American Society of Hematology Annual Meeting and Exposition.
