Cardiff Oncology Reports Positive Phase 2 Data for PLK1 Inhibitor Onvansertib in RAS-Mutated Colorectal Cancer
核心洞察
Cardiff Oncology's Phase 2 CRDF-004 trial showed a 19% improvement in confirmed objective response rate for onvansertib plus standard of care versus control in first-line RAS-mutated metastatic colorectal cancer (搜索).
The 30mg onvansertib dose demonstrated faster time to response, deeper tumor regression, and early signs of progression-free survival benefit with favorable tolerability.
The company maintains $60.6 million in cash resources with projected runway into Q1 2027, positioning for continued development of the PLK1 (搜索) inhibitor.
Cardiff Oncology announced positive interim results from its Phase 2 CRDF-004 trial evaluating onvansertib, a PLK1 (搜索) inhibitor, in combination with standard of care for first-line treatment of RAS-mutated metastatic colorectal cancer (搜索). The data, presented as part of the company's third quarter 2025 financial results, demonstrated meaningful clinical activity in a patient population with significant unmet medical need.
Phase 2 Trial Results Show Promise
As of the July 8, 2025 data cutoff, the Phase 2 CRDF-004 randomized trial demonstrated a 19% improvement in confirmed objective response rate in the 30mg onvansertib arm compared to the control arm in the intent-to-treat population. The study evaluated two doses of onvansertib in combination with standard of care therapeutics.
"This quarter was marked by highly encouraging data from our ongoing CRDF-004 trial evaluating onvansertib in combination with standard of care for first-line RAS-mutated mCRC," said Mark Erlander, Ph.D., Chief Executive Officer of Cardiff Oncology. "At the July 8, 2025 data cutoff, the 30mg onvansertib cohort demonstrated a 19% improvement in confirmed ORR, faster time to response, deeper tumor regression, and early signs of separation in the progression-free survival curves when compared to standard of care alone."
While median progression-free survival has not yet been reached, early PFS data showed a trend favoring the 30mg onvansertib arm versus control. The trial demonstrated dose-dependent responses across all endpoints, including objective response rate, progression-free survival, early tumor shrinkage, and depth of response.
Safety Profile Remains Favorable
Onvansertib continued to be well-tolerated throughout the study period, with no major or unexpected toxicities observed. This safety profile supports the continued development of the PLK1 (搜索) inhibitor in combination with standard therapies.
Addressing Significant Medical Need
The results address a substantial unmet medical need in oncology. According to Dr. Erlander, approximately 150,000 new colorectal cancer patients are diagnosed annually in the U.S. alone. Current standard of care treatments for RAS-mutated metastatic colorectal cancer (搜索) provide median progression-free survival of less than 12 months, with few promising therapies in development for this patient population.
"Onvansertib is uniquely positioned to address a significant medical need and commercial opportunity," continued Dr. Erlander. "With median progression-free survival of less than 12 months on standard of care and few promising therapies in development for RAS-mutated mCRC, we are optimistic that onvansertib has the potential to redefine first-line care for patients."
Development Timeline and Financial Position
Cardiff Oncology expects to report an updated analysis from the ongoing Phase 2 CRDF-004 trial in the first quarter of 2026. This update will provide more mature duration of response and progression-free survival data while continuing to monitor the drug's tolerability profile.
The company maintains a strong financial position to support continued development, with approximately $60.6 million in cash, cash equivalents, and short-term investments as of September 30, 2025. Based on current projections, Cardiff Oncology believes its cash resources are sufficient to fund operations into the first quarter of 2027.
PLK1 Inhibition Strategy
Cardiff Oncology is leveraging PLK1 (搜索) inhibition, described as a well-validated oncology drug target, to develop novel therapies across multiple cancer types. Beyond the colorectal cancer program, onvansertib is being evaluated in ongoing and planned investigator-initiated trials in metastatic pancreatic ductal adenocarcinoma (搜索), small cell lung cancer (搜索), and metastatic triple negative breast cancer (搜索).
The company's development strategy focuses on targeting tumor vulnerabilities to overcome treatment resistance and deliver superior clinical benefit compared to standard of care alone.
