Centessa Pharmaceuticals has announced a strategic leadership transition and significant pipeline advances as the clinical-stage company positions itself as a leader in orexin receptor 2 (搜索) (OX2R (搜索)) agonist therapeutics. The company appointed Mario Alberto Accardi PhD as Chief Executive Officer effective January 1, 2026, succeeding Saurabh Saha MD PhD, who will transition to an advisory role.
Leadership Transition Reflects Strategic Focus
The leadership change underscores Centessa's evolution from a diversified early-stage R&D company to an organization focused exclusively on orexin therapeutics. "This strategic focus has enabled a robust pipeline of potential best-in-class OX2R (搜索) agonists for a broad range of neurological, neurodegenerative and neuropsychiatric disorders," said Francesco De Rubertis PhD, Chair of Centessa's Board.
Dr. Accardi, who previously served as President and founder of Centessa's Orexin Program, brings extensive experience in orexin biology. He was founder and CEO of Orexia Therapeutics (搜索), which successfully raised €40M in Series A funding before its acquisition by Centessa in 2021. ORX750 Shows Promise Across Sleep-Wake Disorders
Centessa's lead candidate ORX750 has demonstrated encouraging results in Phase 2a trials across multiple sleep-wake disorders. In narcolepsy type 1 (搜索) (NT1), the 1.5 mg dose showed greater than 20 minutes change from baseline on the Maintenance of Wakefulness Test (MWT) and reduced Epworth Sleepiness Scale (ESS) scores from 18 to 5. The compound also achieved what management described as "best in class" weekly cataplexy results, including an incidence rate ratio of 0.13 and an 87% reduction.
For narcolepsy type 2 (搜索) (NT2), ORX750 at 4 mg demonstrated greater than 10-minute change from baseline on the MWT and reduced ESS to 8, representing normalization of sleepiness scores. Management noted that no approved drug in NT2 has shown a greater than 10-minute change from baseline on the MWT.
In idiopathic hypersomnia (搜索) (IH), ORX750 showed "statistically and clinically meaningful impact" on multiple efficacy measures, including IH-specific symptoms such as sleep inertia and brain fog as measured by the Idiopathic Hypersomnia Severity Scale (IHSS).
Expanding Into Neuropsychiatric Indications
New preclinical data presented at the 64th Annual Meeting of the American College of Neuropsychopharmacology (ACNP) supports expansion of the orexin platform beyond sleep disorders. The research showed that OX2R (搜索) activation with a highly potent and selective OX2R agonist reduced behavioral despair and enhanced wakefulness in an established animal model of major depressive disorder (搜索).
"These compelling preclinical findings reinforce our strategy of targeting the orexin pathway for the development of new therapeutic options with the potential to address mood symptoms, excessive daytime sleepiness (搜索) (EDS), impaired attention, cognitive deficits, fatigue and other symptoms across multiple neuroscience indications," Dr. Accardi explained.
Pipeline Advancement and Commercial Preparation
The company expects to initiate registrational programs for ORX750 in Q1 2026, with dose selection being a critical consideration. Management is exploring once-daily versus split-dosing regimens and balancing longer duration of action against tolerability concerns such as insomnia at extended exposures.
Centessa plans to advance its next-generation asset ORX142 into clinical trials this quarter, targeting neurological, neurodegenerative and neuropsychiatric disorders. The company is also developing ORX489 for similar indications as part of its multi-asset orexin strategy.
As the company transitions toward a pre-commercial stance, it plans commercial hires for 2026, positioning 2026 as what management describes as "transformational" for the organization.
