Chugai Licenses RAF-MEK Molecular Glue SPYK04 to InxMed in Worldwide Deal
核心洞察
Chugai Pharmaceutical and InxMed (搜索) have signed a worldwide license agreement covering SPYK04, a RAF (搜索)-MEK (搜索) molecular glue discovered by Chugai for the treatment of solid tumors (搜索).
Under the deal, InxMed (搜索) gains exclusive worldwide rights to develop, manufacture and commercialize SPYK04 across all indications, with Chugai receiving an upfront payment and sales royalties.
SPYK04 is a small-molecule compound that binds RAF (搜索) and MEK (搜索), inducing an inactive RAF-MEK complex that inhibits proliferative signaling in cancer cells.
Chugai Pharmaceutical Co., Ltd. and InxMed (搜索) (Hong Kong) Limited announced on September 29, 2026 that they have entered into a worldwide license agreement for SPYK04, a RAF (搜索)-MEK (搜索) molecular glue discovered by Chugai for the treatment of solid tumors (搜索).
Under the terms of the agreement, Chugai will grant InxMed (搜索) an exclusive worldwide license to develop, manufacture and commercialize SPYK04 for all indications. In return, Chugai will receive an upfront payment as well as royalties based on sales following product launch. In addition, if InxMed receives income other than royalties from sublicensees, Chugai will be entitled to receive a certain portion of that income.
A Molecular Glue Approach to RAF-MEK Signaling
SPYK04 is a small-molecule compound discovered by Chugai for the treatment of solid tumors (搜索). It acts as a "molecular glue" that binds RAF (搜索) and MEK (搜索), proteins involved in cancer cell proliferation. By inducing the formation of an inactive RAF-MEK complex, SPYK04 inhibits proliferative signaling in cancer cells and is expected to exert antitumor effects.
The agreement covers development, manufacturing and commercialization across all indications, with InxMed (搜索) taking responsibility for advancing the asset globally.
Company Perspectives on the Collaboration
Chugai's President and CEO, Dr. Osamu Okuda, framed the deal around InxMed (搜索)'s oncology capabilities. "We are pleased to have entered into a license agreement for SPYK04 with InxMed, which has strong expertise in the oncology field," he said. "We expect that InxMed will steadily advance development leveraging the unique characteristics of SPYK04, and we hope that it will be delivered as a cancer treatment to patients around the world as soon as possible."
Dr. Zaiqi Wang, Founder, Chairman and CEO of InxMed (搜索), described the agreement as aligned with the company's precision oncology strategy. "We are thrilled to enter into this licensing agreement with Chugai for asset SPYK04," he said. "This collaboration marks a significant milestone in our mission to deliver further precision oncology therapies. We look forward to rapidly advancing this asset to bring more effective, durable treatment options to cancer patients worldwide."
InxMed's Pipeline and Focus on Tumor Defense
Founded in 2017, InxMed (搜索) is a late clinical-stage biotechnology company dedicated to tackling drug resistance caused by tumor defense mechanisms. Its strategy focuses on critical signaling hubs shared across multiple tumor types, most notably the focal adhesion kinase (FAK) and integrin pathways, which play pivotal roles in tumor cell survival and contribute to treatment failure across various therapeutic modalities. In parallel, the company targets cancer-associated fibroblasts (CAFs) to dismantle the protective barrier around tumor cells.
InxMed (搜索)'s lead program, ifebemtinib, is currently undergoing registrational studies and has been granted Breakthrough Therapy Designation by the China National Medical Products Administration (NMPA) as well as Fast Track Designation by the U.S. FDA. Several additional preclinical candidates that demonstrate synergy with the company's clinical-stage assets are advancing toward clinical studies.
