Expanded Manufacturing Capabilities
The new 50,000-square-foot center houses 12 state-of-the-art clean rooms designed to prevent contamination during the production of gene and cell therapies, making it one of the largest academic centers of its kind worldwide. The facility was built with extraordinary attention to detail, featuring air filtration systems cleaner than hospital operating rooms and tightly controlled temperature and humidity at the individual room level.
"What makes the Applied Gene and Cell Therapy Center unique is that it's not just a research lab and it's not just a manufacturing facility – it's both," said Chaozhong "Charles" Zou, executive director of the center. "The center is tightly connected to the clinical side of Cincinnati Children's, which means discoveries can move faster from bench to bedside, enabling patients to gain access to new therapies sooner."
Clinical Impact and Treatment Focus
The center develops vectors and cellular products for various conditions, including telomere biology disorders resulting from gene mutations that can lead to premature aging, organ problems, and predisposition to cancers such as leukemia. Other therapeutic targets include sickle cell disease, which causes severe pain and organ damage, and hereditary protein alveolar proteinosis, a lung disease causing shortness of breath, chronic cough, and fatigue.
Dr. Stella Davies from the Cancer & Blood Diseases Institute emphasized the transformative potential of these treatments: "What we can do here in this facility is generate new treatments that don't use chemotherapy... they use cells and modified DNA to cure children without side effects." Dr. Kasiani Myers from the Division of Bone Marrow Transplantation added, "We've seen it change outcomes in patients with cancer diagnoses."
Investment and Timeline
Cincinnati Children's invested $60 million in the new facility, including design, construction, and equipment. The building, previously an auto parts distribution facility, encompasses 111,000 square feet on a 14.6-acre site. The center will undergo a one-year certification process to meet requirements for manufacturing gene, cell, and immunotherapy products for clinical trials, with onsite production expected to begin by early 2027.
Up to 100 Cincinnati Children's employees will work at the expanded center, including 24 newly recruited positions. Approximately half of the new hires are scientists, including biologists and quality assurance specialists focused on Current Good Manufacturing Practice (cGMP) standards.
Addressing Global Medical Needs
The expansion addresses significant medical needs, with over 300,000 children worldwide diagnosed with cancer annually and an estimated 8 million born with birth defects. Cincinnati Children's, which ranks No. 1 nationally for pediatric cancer care, serves patients from all 50 states and dozens of countries seeking treatment for complex or rare disorders.
"Our Applied Gene and Cell Therapy Center is the launchpad for new cures," said Dr. Tina Cheng, director of the Cincinnati Children's Research Foundation and chief medical officer. "This center takes promising science, makes it real, and helps bring hope to patients and families who need it most."
The facility builds on nearly 25 years of success from the previous center, formerly known as the Cincinnati Children's Translational Core Laboratory, which has been a leader in developing, testing, and manufacturing drugs and therapeutics for early-phase clinical trials. The new center is designed to enhance collaboration with pharmaceutical companies, biotech firms, universities, and other health systems to accelerate therapy development.
