Congruence Therapeutics (搜索) and Ono Pharmaceutical Co., Ltd. (搜索) announced the expansion of their existing multi-target research partnership through a new collaboration agreement covering two additional programs in neurology and immunology. The expanded partnership, announced March 2, 2026, builds on their original collaboration that began in December 2024.
Partnership Structure and Financial Terms
Under the new agreement, Congruence will lead efforts to discover small molecule modulators (搜索) targeting two clinically validated high-value targets in neurology and immunology, advancing them to the development candidate stage. Ono will have the option to secure exclusive worldwide rights to further develop and commercialize development candidates generated under the agreement.
Congruence will receive an undisclosed upfront payment and is eligible for additional payments upon achievement of milestones in discovery, development, approval and commercialization, plus tiered royalties based on annual net sales of related products. Ono will also reimburse Congruence for internal and external research costs incurred in connection with the expanded partnership activities.
Revenir™ Platform Technology
The collaboration continues to leverage Congruence's proprietary computational drug discovery engine, Revenir™ (搜索), which systematically captures the biophysical features of proteins in different functional states to enable a highly differentiated approach to small-molecule discovery. The platform examines surface features and biophysical descriptors across protein conformers to predict small molecule induced modulation of physiologic protein states.

Clinical Pipeline Progress
Congruence's wholly owned program directed to MC4R-deficient genetic obesity (搜索) is entering a Phase 1/1b clinical study this month. The company will also nominate development candidates in the coming weeks for programs targeting GBA1-driven Parkinson's Disease (搜索) and Alpha-1 Antitrypsin deficiency (搜索).
The original collaboration between the companies focuses on leveraging Revenir™ (搜索) to discover novel small molecule correctors (搜索) directed to mutants of a difficult-to-drug cancer (搜索) target.
