Cue Biopharma and ImmunoScape (搜索) have announced a strategic collaboration to advance a novel "Seed-and-Boost" immunotherapy approach that could transform cell therapy treatment for solid tumors (搜索). The partnership combines Cue Biopharma's clinically-validated Immuno-STAT (搜索) T-cell engagers with ImmunoScape's proprietary T cell receptor (搜索) (TCR (搜索)) technology to overcome core limitations of existing cell therapies.
Revolutionary Seed-and-Boost Mechanism
The innovative therapeutic approach involves a two-step process designed to enhance T cell therapy efficacy while simplifying manufacturing. First, a minimal starting dose of ImmunoScape (搜索)'s tumor-specific TCR (搜索)-engineered T cells (TCR-Ts) are administered to patients as the "Seed" component. This is followed by administration of Cue Biopharma's TCR-matched IL-2 Immuno-STAT (搜索) molecules as the "Boost" component.
The TCR (搜索)-selective engagement enables targeted expansion and activation of T cells directly within the patient while avoiding systemic immune activation. This off-the-shelf Immuno-STAT (搜索) dosing allows for precise in vivo modulation of the tumor-specific T cell response to enhance efficacy, persistence and tumor infiltration while mitigating T cell exhaustion.
"By requiring only small starting numbers of engineered T cells, this strategy simplifies manufacturing and aims to deliver deeper and durable clinical efficacy for patients along with improved tolerability and quality of life," according to the companies' joint announcement.
Clinical Validation and Preclinical Success
The CUE-100 series represents a novel class of injectable biologics that leverage TCR (搜索) specificity to enable selective engagement and activation of targeted disease-specific T cells. The core protein framework facilitates TCR engagement while delivering Interleukin-2 (IL-2) as a key costimulatory molecule for selective T cell activation and expansion.
Phase 1 clinical datasets from the CUE-100 series programs have demonstrated clinical activity in several metastatic cancers (搜索) without the significant toxicities often caused by traditional immune-activating IL-2 delivery. The binding affinity of IL-2 for its receptor has been deliberately attenuated to achieve preferential selective activation of tumor-specific effector T cells while reducing potential effects on regulatory T cells or broad systemic activation.
Compelling preclinical data generated in multiple solid tumor models, including pancreatic and ovarian cancers, demonstrates the clinical potential of this novel immunotherapy approach. IND-enabling studies are on track for 2027 submission.
Strategic Partnership Terms
Under the collaboration agreement, Cue Biopharma is entitled to receive an upfront total payment of $15 million, with $10 million in Q4 2025 and $5 million in November 2026, as well as a 40% equity stake in ImmunoScape (搜索). Additionally, Cue Biopharma is eligible for high-single-digit royalty payments on net sales.

The company's proprietary Deep Immunomics technology platform enables highly sensitive, large-scale mining and immune profiling of T cells in cancer patient samples to identify novel, therapeutically relevant TCRs.
