CytoAgents (搜索), Inc., a clinical-stage biotechnology company, will present preclinical data demonstrating that its investigational drug CTO1681 can reduce dangerous inflammatory side effects of CAR-T cell therapy without compromising the treatment's cancer-fighting effectiveness. The data will be presented at the European Society for Medical Oncology (ESMO) Immuno-Oncology Congress 2025, taking place December 10-12, 2025 in London.
Novel Approach to CAR-T Safety
CTO1681 is an orally administered immune modulator designed as a steroid-sparing inhibitor of prostaglandin (搜索)-mediated inflammation. The drug specifically targets inflammation in the tumor microenvironment and aims to mitigate cell therapy-induced toxicities frequently observed in lymphoma (搜索) patients receiving CAR-T cell therapy.
The company is currently enrolling patients in a Phase 1b/2a clinical trial evaluating CTO1681 in lymphoma (搜索) patients receiving CAR-T cell therapy who are at risk for inflammatory-driven toxicities.
Preclinical Data Shows Promise
At ESMO, CytoAgents (搜索) will present findings from two non-clinical studies that demonstrate CTO1681's ability to attenuate key cytokines known to drive Cytokine Release Syndrome (搜索) (CRS) without compromising CAR-T cell-mediated tumor killing both in vitro and in vivo.
The studies show that CTO1681 does not interfere with CD19 (搜索) CAR-T cell expansion and anti-tumor activity in vivo while reducing the inflammatory cytokines associated with CRS and immune cell-associated neurotoxicity syndrome (搜索) (ICANS).
