Debiopharm Advances Bispecific ADC Development with SunRock Biopharma's Anti-HER2/HER3 Antibody License
核心洞察
Debiopharm (搜索) has licensed SunRock Biopharma (搜索)'s SRB21, a bispecific antibody targeting both HER2 (搜索) and HER3 (搜索) receptors, to develop the ADC Debio 2512 (搜索) using their proprietary MultiLINK™ Linker Technology.
The dual-targeting approach aims to overcome resistance mechanisms that limit current HER2 (搜索)-directed therapies by simultaneously targeting HER2 and HER3 (搜索) receptors in aggressive cancer types.
HER2-driven cancers (搜索) are characterized by faster growth, earlier relapse, and higher treatment resistance, with many patients ultimately developing resistance and metastasis despite recent therapeutic advances.
Debiopharm (搜索), a Swiss-based biopharmaceutical company, has exercised its option to license SunRock Biopharma (搜索)'s bispecific antibody SRB21, which targets both HER2 (搜索) and HER3 (搜索) human epidermal growth factor receptors. The licensing agreement, initially announced in 2023, will combine this antibody with Debiopharm's proprietary MultiLINK™ Linker Technology to develop a new antibody-drug conjugate (ADC) designated Debio 2512 (搜索).
Dual-Targeting Strategy for Enhanced Cancer Treatment
The collaboration leverages Debiopharm (搜索)'s "Trifecta" approach, utilizing the bispecific antibody to create a fit-for-purpose ADC through proprietary linkers and smart payload selection. Bispecific ADCs target multiple antigens simultaneously, potentially offering higher efficacy and specificity while reducing off-target effects and overcoming resistance mechanisms.
"Bi-specific ADCs hold significant promise as the next generation of cancer therapies," stated Bertrand Ducrey, CEO of Debiopharm (搜索). "We're excited to confirm this licensing option as it allows us to apply our MultilinkTM Linker Technology to a dual-targeted design that could pave the way for breakthrough therapies in HER-driven cancers."
Addressing Treatment Resistance in Aggressive Cancers
HER2-driven cancers (搜索) are widely recognized as more aggressive forms of disease, marked by faster growth, earlier relapse, and a higher likelihood of treatment resistance. While recent therapies have provided meaningful advances, many patients ultimately develop resistance and metastasis, limiting the durability of response and long-term survival benefits.
The dual-targeting strategy may allow for more potent delivery of cytotoxic payloads directly to cancer cells while sparing healthy tissues, offering breakthrough options for patients with resistant cancer types. By targeting simultaneously HER2 (搜索) and HER3 (搜索), this approach has the potential to overcome resistance mechanisms that limit current HER2-directed therapies.
Competitive Landscape and Market Position
A review of the oncology pipeline reveals only a handful of other HER2 (搜索) x HER3 (搜索)-targeting projects, with Merus's MAb Bizengri being the most notable. Although Bizengri received FDA accelerated approval in December for second-line, NRG1 fusion-positive pancreatic cancer (搜索) and NSCLC (搜索), Merus subsequently sold commercial rights to Partner Therapeutics for an undisclosed fee.
Debiopharm (搜索) appears to envision broader applications for Debio 2512 (搜索), potentially targeting the same indications as established HER2 (搜索) ADCs like AstraZeneca and Daiichi (搜索)'s Enhertu, which generated $3.8 billion in sales in 2024. The company is also developing a preclinical anti-HER3 (搜索) ADC, Debio 0532, though challenges with similar approaches have emerged in the field.
Partnership Background
SunRock Biopharma (搜索), a Galician company supported by the regional government Xunta de Galicia through Xesgalicia, focuses on developing antibodies against highly invasive tumors with urgent clinical needs in oncology. The two companies have been collaborating on ADCs since September 2023, when HER2 (搜索) and HER3 (搜索) were initially identified as potential targets. Financial terms of the current licensing agreement were not disclosed.
