Denali Therapeutics Secures $275 Million Royalty Deal with Royalty Pharma for Hunter Syndrome Therapy

TransportVehicle Platform Enables Brain Penetration
Tividenofusp alfa is an enzyme replacement therapy enabled by Denali's proprietary TransportVehicle platform, designed to deliver therapeutics across the blood-brain barrier (搜索). A Biologics License Application (BLA) for accelerated approval is currently under review by the U.S. Food and Drug Administration (FDA) with a Prescription Drug User Fee Act (PDUFA) target date of April 5, 2026.

Addressing Unmet Medical Need in Hunter Syndrome

Financial Structure and Milestones
The transaction is contingent upon various closing conditions, including Denali achieving U.S. FDA accelerated approval of tividenofusp alfa. Upon closing, Royalty Pharma will make an initial payment of $200 million, with an additional $75 million payment due upon achieving European Medicines Agency (EMA) approval by December 31, 2029.
In exchange for the funding, Royalty Pharma will receive a 9.25% royalty on worldwide net sales of tividenofusp alfa. The royalty payments will terminate upon reaching a multiple of 3.0x of the total investment, or 2.5x if achieved by the first quarter of 2039.
