DOC1021 represents a first-in-class, patient-derived double-loaded dendritic cell therapy that combines tumor lysate and amplified tumor-derived mRNA. According to Jay Hartenbach, President and COO of Diakonos Oncology, "These clinical data continue to validate DOC1021's novel mechanism, which leverages a patient's full complement of tumor antigens (搜索) to drive robust immune responses against malignancy."
The therapy's unique double-loading approach mimics viral infection physiologically, creating what the company describes as a synergistic and exponentially more powerful tumor killing response. This mechanism permits complete targeting of the total cancer antigen pool without requiring molecular modification or genetic engineering of the patient's immune cells.
Clinical Trial Updates Across Multiple Indications
The AACR presentation, titled "Clinical and Immunologic Assessment of DOC1021 Dendritic Cell Therapy in Resectable or Borderline Resectable Pancreatic Cancer (搜索)," will provide updated data from the ongoing pancreatic cancer study. The poster will be presented on Monday, April 20, from 9:00 AM–12:00 PM PT as part of the Phase I Clinical Trials in Progress session.
At AAN, researchers will present "DOC1021 Cell-based Immunotherapy in Combination with Standard Chemoradiation for Adjuvant Therapy of Glioblastoma (搜索): Early Results from an Expanded Access Protocol of a Phase I Trial." This marks the first report of results from the expanded access program in glioblastoma and will be presented on Wednesday, April 22, from 11:45 AM–12:45 PM CT.
Regulatory Recognition and Development Pipeline
DOC1021 has garnered significant regulatory attention, receiving FDA Fast Track designation for both the glioblastoma (搜索) program in October 2023 and the pancreatic cancer (搜索) program in May 2024. The therapy also received Orphan Drug Designation for the glioblastoma program in January 2024.
