Novel Dual-Targeting Mechanism
DNTH212 (搜索) represents an innovative approach to autoimmune treatment by simultaneously targeting two clinically validated pathways. The investigational therapy is an extended half-life bifunctional fusion protein that targets plasmacytoid dendritic cell (pDC) BDCA2 (搜索) to reduce Type 1 interferon production while simultaneously inhibiting BAFF/APRIL (搜索) to suppress B cell function.
"By targeting both the innate and adaptive immune systems via two clinically validated pathways that are known drivers of autoimmune disease pathogenesis, this complementary and differentiated approach has the potential to address multiple autoimmune indications with improved outcomes," according to the company's announcement.
Plasmacytoid dendritic cells are specialized immune cells that produce large amounts of Type 1 interferons (IFN-α and IFN-β). High levels of Type 1 interferon drive chronic inflammation and tissue damage, making Type 1 interferon inhibition a promising therapeutic strategy in multiple autoimmune diseases (搜索). Meanwhile, BAFF/APRIL (搜索) activation of B cells generates autoantibodies against tissues that trigger inflammation and tissue damage.
Superior Preclinical Performance
In preclinical studies, DNTH212 (搜索) demonstrated superior performance compared to existing therapies targeting similar pathways. The therapy showed comparable IFN-α inhibition and superior pDC depletion in vitro compared to litifilimab, a late-stage clinically validated BDCA2 (搜索) monoclonal antibody. Additionally, DNTH212 demonstrated superior inhibition of immunoglobulins (IgM, IgA, and IgG) in non-human primates compared to povetacicept, a late-stage clinically validated BAFF/APRIL (搜索) inhibitor.

Clinical Development Timeline
DNTH212 (搜索) has already received FDA IND clearance in September 2025 and is expected to clear regulatory approval in China in Q4 2025. A two-part Phase 1 study in China is planned to initiate by year-end 2025, with Part A involving healthy volunteers and Part B focusing on patients with systemic lupus erythematosus (搜索). Top-line results in healthy volunteers are expected in the second half of 2026.
The therapy is designed for patient-friendly subcutaneous self-administration with dosing every four weeks or less frequently, potentially positioning it as a first-line biologic treatment across multiple autoimmune disorders.
Financial Terms and Strategic Impact
Under the licensing agreement terms, Dianthus will pay Leads Biolabs (搜索) up to $38 million in initial payments, comprising $30 million in upfront and near-term milestone payments plus an additional $8 million milestone upon initiation of a Dianthus-led Phase 1 study. Leads Biolabs remains eligible for up to an additional $962 million in total development, regulatory approval, and sales-based milestones across multiple indications, as well as tiered royalties from mid-single digits up to low double-digit percentages on ex-Greater China net sales.
Following the licensing agreement, Dianthus will maintain an estimated pro forma cash balance of approximately $525 million. This includes preliminary and unaudited cash, cash equivalents, and investments as of September 30, 2025, of approximately $555 million, less the $30 million in upfront and near-term milestone payments. The company has reaffirmed its cash runway guidance extending into 2028, including planned development of DNTH212 (搜索).
