The trial specifically enrolled the 10% of cystic fibrosis (搜索) patients who are either ineligible for or do not receive CFTR (搜索) modulator therapies, representing those for whom existing treatments are least effective. This patient population faces significant unmet medical need, as CFTR modulators (搜索), while transformative for many cystic fibrosis patients, leave a meaningful proportion without effective treatment options.
Novel Mechanism Shows Promise
ETD001 represents the first ENaC (搜索) blocker to demonstrate efficacy in cystic fibrosis (搜索) patients. The drug is designed to increase lung function by improving mucus clearance and reducing airway obstruction through its differentiated mechanism of action. Exploratory analysis revealed that participants had a three times higher likelihood of improving ppFEV1 when receiving ETD001 compared to placebo.

Trial Design and Safety Profile
The Phase 2 study was conducted across multiple sites in the UK, Germany, France, and Italy using a two-part design. Part A assessed safety and tolerability of repeat inhaled doses over 7 days, while Part B employed a double-blind, placebo-controlled, crossover design examining the effects of twice-daily dosing of ETD001 (4.5 mg) over 28 days. Participants were randomized in a two-group, two-sequence crossover design with a 28-day washout period.
The safety profile showed ETD001 was overall well-tolerated in cystic fibrosis (搜索) patients, with adverse events consistent with those expected in this patient population receiving inhaled medicines. Expanding Development Strategy
Enterprise Therapeutics (搜索) plans to advance ETD001 into longer-duration Phase 2b dose-ranging trials and evaluate its efficacy in combination with CFTR (搜索) modulator therapies. The company is also exploring the drug's potential in other muco-obstructive lung diseases, including non-CF bronchiectasis (搜索), where patients similarly suffer from high mucus burden and mucus plugging.
"We are encouraged by the potential of ETD001 to be a novel therapeutic option for improving the lives of all pwCF, particularly those currently without effective therapies," said Dr. Gupta. The results will be presented at the European Cystic Fibrosis (搜索) Society conference in Lisbon in June.
Addressing Critical Medical Need
Cystic fibrosis (搜索) affects over 100,000 people worldwide and leads to reduced life expectancy due to failed mucociliary clearance and mucus congestion in the lungs, which creates cycles of infection and inflammation and ongoing decline in lung function. While CFTR modulators (搜索) have transformed outcomes for many patients, a significant gap remains for those who cannot benefit from these therapies.
