Novel Mechanism of Action
Verdinexor tablets target a unique cellular mechanism involved in cancer (搜索) progression. Cancer cells can disable normal cellular growth regulation by driving tumor-suppressing proteins (搜索) out of the nucleus. The drug works by blocking this exit route, allowing these tumor-suppressing proteins to remain normally active within the cell nucleus, thereby helping control the growth and prevent the spread of lymphoma (搜索) cells. According to the FDA, verdinexor tablets is selectively cytotoxic for tumor cells.
Clinical Efficacy Data
In a clinical field study involving 160 dogs with B-cell or T-cell lymphoma (搜索), verdinexor tablets demonstrated significant clinical benefits. Dogs treated with the drug experienced a significantly longer time to disease progression compared to the control group—37 days versus 23 days (p = 0.011). The study included dogs with both newly diagnosed and relapsed lymphoma (搜索), with 134 dogs included in the effectiveness analysis.
Oncologists have observed particularly promising results with T-cell lymphoma (搜索) cases. "Oncologists have observed the drug has a very high response rate against T-cell lymphoma," noted Craig A. Clifford, DVM, MS, DACVIM (Oncology). Additionally, the drug has not been shown to cause resistance to other chemotherapy types, according to Pamela D. Jones, DVM, DACVIM (Oncology), DACVR (Radiation Oncology).
Administration and Dosing
Verdinexor tablets are available only by prescription from a licensed veterinarian, with dog owners administering the medication orally at home following veterinary instructions. The drug is administered twice per week with at least 72 hours between doses, at a starting dose of 1.25 mg/kg. If the medication is tolerated after 2 weeks, the dose can be increased to 1.5 mg/kg, maintaining the 72-hour interval between doses.
Each dose must be given with food or immediately after a meal, as this increases the amount of drug absorbed into the bloodstream, according to the FDA.
Regulatory Pathway and Market Impact
The full approval follows four annual renewals of the FDA's conditional approval, during which Anivive Lifesciences (搜索) generated the necessary effectiveness data. The FDA first granted conditional approval to verdinexor tablets in January 2021 through the agency's Minor Use/Minor Species pathway, a program designed to incentivize development of drugs for uncommon conditions in major species.
According to the FDA, fewer than 80,000 dogs are affected by canine lymphoma (搜索) each year, qualifying verdinexor tablets as a minor use in a major species under this regulatory framework.
Clinical Significance
The approval addresses a significant gap in canine lymphoma (搜索) treatment options. "It's not meant to be a silver bullet, a cure-all for lymphoma," said Clifford, "but to me, knowing that 80% of lymphomas are left untreated, this gives the primary care [practitioner] a game changer—something else that they can utilize, other than just prednisone."
Clifford emphasized the drug's potential to expand treatment access: "We're expanding access, meaning more cases are going to get treated, and it's an unmet need, and I think this drug checks both of those boxes."
