Genentech Expands Holly Springs Manufacturing Investment to $2 Billion for Obesity Drug Production

Genentech, a subsidiary of Swiss pharmaceutical giant Roche Group, announced Tuesday it will more than double its investment in a new North Carolina biomanufacturing facility to approximately $2 billion, marking a significant expansion of the company's first East Coast manufacturing site.
The increased commitment represents a substantial jump from the initial $700 million investment announced in May 2025 for the Holly Springs facility. Construction on the 700,000-square-foot project began in August 2025, with operations expected to commence by 2029.
Focus on Metabolic Disease Production
The state-of-the-art facility will specialize in producing next-generation treatments for metabolic conditions, with a particular emphasis on obesity medications. The expansion will allow Genentech to increase production volume and scale manufacturing capacity within the facility through advanced biomanufacturing technologies, automation, and digital capabilities.
"This additional investment will create more high-quality jobs, strengthen local partnerships, and ensure a resilient supply of medicines for years to come, allowing us to bring life-changing medicines to patients faster and more reliably," said Genentech CEO Ashley Magargee.
Economic Impact and Job Creation
The expanded investment is projected to support more than 2,000 jobs total, including over 500 high-wage manufacturing positions and 1,500 construction jobs. The additional investment will create approximately 100 new jobs beyond the original projections, with manufacturing positions offering an average salary of about $120,000.
Genentech received a $13.1 million state incentive package for the project, including a $9.8 million job creation grant from North Carolina, according to the state's Department of Commerce.

The company cited the highly skilled local workforce, strong academic institutions, and proximity to leading life science companies in the Raleigh-Durham area as key factors in the decision to expand the investment. Holly Springs has emerged as a national biopharmaceutical hub, attracting recent investments from companies including Fujifilm Diosynth Biotechnologies (搜索) and Amgen.

The Holly Springs expansion supports Roche and Genentech's broader $50 billion commitment to U.S. manufacturing and R&D investment. The company's current U.S. footprint includes 13 manufacturing sites and 15 R&D facilities across pharmaceutical and diagnostics divisions, employing approximately 25,000 people at 24 locations in eight states.
The investment aligns with a broader pharmaceutical industry trend toward domestic manufacturing. Johnson & Johnson recently announced plans for two new U.S. manufacturing facilities as part of its $55 billion U.S. investment program, while Novartis announced a $23 billion investment in U.S.-based infrastructure over the next five years.
The expansion reflects pharmaceutical companies' response to supply chain disruptions and policy priorities aimed at reshoring pharmaceutical manufacturing to strengthen domestic innovation and production capabilities.
